摘要
目的研究黑柴胡提取物乙酸乙酯萃取部位的化学成分。方法使用75%乙醇对黑柴胡干燥根进行提取,所得浸膏在水中混悬,用石油醚、乙酸乙酯、正丁醇依次萃取。其中乙酸乙酯萃取部位使用硅胶柱、凝胶柱及制备液相等色谱分离手段对其中所含的化合物进行分离纯化,化合物结构通过理化常数对照和核磁共振、质谱等波谱学技术得以鉴定。结果从黑柴胡95%乙醇提取物乙酸乙酯部位中分离鉴定了10个化合物,分别为柴胡皂苷元A(1),柴胡皂苷元C(2),柴胡皂苷元D(3),柴胡皂苷元E(4),柴胡皂苷元F(5),柴胡皂苷元Q(6),3,5,7-三羟基-3’,4’-二甲氧基黄酮(7),山奈酚(8),槲皮素(9),异鼠李素(10)。结论从黑柴胡醇提物乙酸乙酯部位中分离得到6个柴胡皂苷元类化合物和4个黄酮类化合物,其中化合物2~7为首次从该植物中分离得到。
Objective To study the chemical constituents in the ethyl acetate fraction from Bupleurum smithii Wolff.Methods The roots of Bupleurum smithii were extracted with 75%alcohol.The crude extract was then extracted with petroleum ether,ethyl acetate and n-butanol successively.Repeated silica gel column chromatography,Sephadex LH-20 column and preparative HPLC were applied for isolation and purification of the compounds in ethyl acetate fraction.Chemical structures of the compounds were elucidated by spectra analysis such as NMR and ESI-MS as well as physical and chemical properties.Results Ten compounds were isolated from the root of Bupleurum smithii.They were identified as saikogenin A(1),saikogenin C(2),saikogenin D(3),saikogenin E(4),saikogenin F(5),saikogenin Q(6),3,5,7-trihydroxy-3’,4’-dimethoxyflavone(7),kaemferol(8),quercetin(9),isorhamnetin(10).Conclusion Six saikogenins and four flavones were isolated from Bupleurum smithii.Compounds 2-7 were isolated for the first time from this plant.
