摘要
分别制备了黄芪多糖的微乳、水凝胶及基于微乳的凝胶。继而以经微针技术处理的大鼠离体皮肤为屏障,使用Franz扩散池进行体外渗透试验,考察微针技术联合微乳型凝胶对黄芪多糖经皮渗透的影响。所得黄芪多糖微乳平均粒径(31.2±0.8)nm,多分散系数0.168±0.082。体外透皮试验结果显示,对照组(微乳型凝胶,未经微针处理)、水凝胶组(经微针处理)和微乳型凝胶组(经微针处理)的稳态渗透速率分别为50.08、74.08和110.83μg·cm-2·h-1,12 h累积渗透量分别为(601.28±15.76)、(889.00±25.16)和(1330.07±29.06)μg/cm2。可见,在微针技术辅助给药基础上,基于微乳的凝胶能显著促进黄芪多糖的透皮吸收,为透皮给药系统的开发及应用提供了新的方法和思路。
Microemulsion,hydrogel and microemulsion-based gel loaded with Astragalus polysaccharide were prepared,and the transdermal behaviors from two kinds of gel through excised rat skin with microneedle treatment were compared.The transdermal experiment was conducted by Franz diffusion cell,and the effect of microneedle technique combined with microemulsion-based gel on the permeation of Astragalus polysaccharide through rat skin was also investigated.The average particle size and polydispersity(PDI)of the microemulsion were(31.2±0.8)nm and 0.168±0.082.The results of in vitro transdermal test showed that the steady-state permeation rates of the control group(microemulsion-based gel and without microneedle treatment),hydrogel group(with microneedle treatment)and microemulsion-based gel group(with microneedle treatment)were 50.08,74.08 and 110.83μg·cm-2·h-1,respectively.And their cumulative amounts permeated at 12 h were(601.28±15.76),(889.00±25.16)and(1330.07±29.06)μg/cm2,respectively.It was concluded that the microemulsion-based gel could significantly enhance the transdermal permeation of Astragalus polysaccharide on the basis of microneedle-assisted drug delivery.
