摘要
研究阿尔兹海默症(Alzheimer's disease,AD)转基因鼠(5XFAD)认知功能障碍与海马神经节律的关系,探讨海马theta节律在AD发展过程中的变化。设计AD转基因鼠和同源WT鼠在2月龄和5月龄的水迷宫及在体电生理实验,分析对比AD鼠和WT鼠行为学表现及海马theta节律频谱特征。AD鼠和WT鼠在2月龄时行为学表现无差异,AD鼠相较于WT鼠theta节律主频降低;在5月龄时,AD鼠出现明显的运动障碍和认知障碍,theta节律能量降低。结果表明,AD疾病发展过程中,海马神经节律的异常要先于认知功能障碍。研究结果为通过神经调控延缓AD认知障碍提供了实验依据。
To investigate the relationship between cognitive dysfunction of Alzheimer′s disease(AD)transgenic mice(5XFAD)and hippocampal circadian rhythm,and to obtain the change pattern of hippocampal theta rhythm during AD development.Water mazes and in-vivo electrophysiological experiments were designed for two-and five-month-old AD transgenic mice and homologous WT mice,and their behavioral characteristics and the hippocampal theta rhythm spectrum characteristics were analyzed and compared.There was no difference in behavior between AD mice and WT mice at 2 months of age.Compared with WT mice,AD mice had a lower frequency of theta rhythm.At 5 months of age,AD mice showed significant dyskinesia and cognitive impairment,theta rhythm energy decreased.During the development of AD disease,abnormal hippocampal rhythms preceded cognitive dysfunction.The results provide a basis for relieving AD cognitive impairment through neural regulation.
