水杉核糖体蛋白基因MG-RPS6的生物信息学分析

摘要 水杉(Metasequoia glyptostroboides)是中国著名的杉科孑遗植物。近些年来,对水杉的生态习性、病虫害防治和生物活性的研究已经取得了一定的成果,但是在分子水平的相关研究还很少。本研究以水杉原生种为植物材料,从三代测序的全长转录组文库中克隆了一个核糖体蛋白基因(MG-RPS6)。对MG-RPS6基因进行生物信息学分析,结果显示:该基因的全长c DNA为1365 bp,分子量为423.20 kD,编码氨基酸数量为251个,编码蛋白为核糖体蛋白S6;该蛋白的分子量为28609.46 kD,等电点为10.76,总平均吸水值为-0.847,不稳定指数为47.21;该蛋白含有20个常见功能位点,有28个磷酸化位点,含有典型的核糖体蛋白S6e结构域;二级结构包括8个折叠区、3个螺旋区和一些无规则卷曲区;三级结构分析结果进一步证实本研究克隆的基因为真核生物核糖体蛋白S6基因;通过构建系统进化树,证实水杉和日本柳杉具有较近的亲缘关系。本实验结果可以为水杉及其它物种的核糖体蛋白基因的结构和功能研究提供参考。 Metasequoia glyptostroboides is a famous Chinese relict plant.In recent years,some achievements have been made in ecological habits,pest control and biological activation in Metasequoia glyptostroboides,but few studies have been reported at molecular level.In this study,Metasequoia glyptostroboides was used as plant materi al,and its 40 S ribosomal protein gene(named MG-RPS6)was cloned from a full-length transcriptome library based on three-generation sequencing technology.The results of bioinformatics analysis for the MG-RPS6 gene showed that the total length of the gene is 1365 bp,the molecular weight is 423.20 kD,its number of encoded amino acids is 251,and the encoding protein is ribosomal protein S6;The molecular weight of this protein is 28609.46 kD,the isoelectric point is 10.76,the total average water absorption value is-0.847,and the instability index is 47.21;The protein contains 20 common functional sites,28 phosphorylation sites and a typical ribosomal protein S6 e domain;the secondary structure includes 8 folding areas,3 helix areas,and some coil regions;The result of tertiary structure analysis further demonstrated that the protein encoded by MG-RPS6 gene was eukaryotic ribosomal protein S6.The constructed evolution tree revealed the closest relationship between Metasequoia glyptostroboides and Cryp-%tomeria japonica.The experimental results could provide reference for the study of the structure and function of ribosomal protein genes in Metasequoia glyptostroboides and other species.
