HPLC法测定中药制剂芦荟散中芦荟苷的含量

Determination of aloin in traditional Chinese medicine aloe powder by HPLC

目的采用HPLC法测定中药制剂芦荟散中芦荟苷的含量。方法采用HPLC法,条件为色谱柱:ACE C18(4.6mm×250mm,5μm);流动相:乙腈-水(25∶75);柱温:30℃;波长:355nm;流速:1.0mL/min;进样量:10μL。进行方法学线性、重复性、回收率、耐用性、稳定性及检出限的考察。结果芦荟苷在0.04~0.25mg/mL浓度范围内与峰面积呈良好的线性关系,回归方程为y=192.84x+1.2883,R2=0.9996;重复性RSD为1.56%,重复性良好;回收率平均值为96.52%,RSD为0.92%,回收率良好;耐用性考察不同流速、不同柱温、不同色谱柱条件下样品的含量,RSD分别为0.79%、0.17%、0.57%,耐用性良好;芦荟苷的检出限为0.01μg/mL。结论建立的方法准确可靠,重复性好,分离度高,灵敏度高。

Objective To determine the content of aloin in traditional Chinese medicine preparation aloe powder by HPLC.Methods HPLC method was used,and the conditions were as follows:chromatography column,ACE C18(4.6mm×250mm,5μm);Mobile phase,acetonitrile-water(25:75);Column temperature,30℃;Wavelength,355nm;flow rate,1.0mL/min;Injection volume,10μL.The methodological linearity,repeatability,recovery rate,durability,stability,and detection limits were detected.Results Aloin had a good linear relationship with the peak area in the concentration range of 0.04 to 0.25mg/mL.The regression equation was y=192.84x+1.2883,R2=0.9996;The repeatable RSD was 1.56%,and the repeatability was good;the average value of repeatability was 96.52%,the RSD was 0.92%,and the recovery rate was good.When examining the durability,the content of the sample under different flow rates,different column temperatures,and different column conditions,the RSD was 0.79%,0.17%,and 0.57%,respectively,and the durability was good;the detection limit of aloin was 0.01μg/mL.Conclusion The established method is accurate and reliable,with good reproducibility,high resolution and high sensitivity.
