摘要
目的:探讨EGCG对体外培养宫颈癌HeLa细胞增殖、凋亡和迁移的影响。方法:通过MTT法、流式细胞法、细胞划痕法、Transwell法及蛋白质免疫印迹法对宫颈癌细胞增殖、周期、凋亡进行检测分析。结果:未经药物处理时,空白对照组细胞凋亡率相对较低,EGCG为0mg/L凋亡率仅为(0.7±0.1)%,G1为(44.2±2.1)%,S为(43.3±1.3)%,M为(7.5±1.3)%,而EGCG为40mg/L时,凋亡率仅为(17.4±1.7)%,G1为(58.3±2.5)%,S为(32.7±2.6)%,M为(6.3±1.4)%;当EGCG能促进细胞凋亡,EGCG浓度越高,凋亡率越高,G1细胞比例增加,S期细胞比例相应下降,EGCG剂量越大,G1细胞及S期细胞增大、下降比例越大。结论:EGCG对体外培养可对宫颈癌HeLa细胞增殖产生抑制作用,其迁移、凋亡作用也相对减退。
Objective:To investigate the effect of Epigallocatechin gallate(EGCG)on the proliferation,apop-tosis and migration of HeLa cells of cervical cancer cultured in vitro.Methods:The proliferation,cycle and apopto-sis of cervical cancer cells were detected by MTT,flow cytometry,cell scratch,Transwell and western blot.Re-sults:Without drug treatment,the apoptosis rate of blank control group was relatively low.When EGCG was 0mg/L,the apoptosis rate was only(0.7±0.1)%,G1(44.2±2.1)%,S(43.3±1.3)%,and M(7.5±1.3)%.When EGCG was 40mg/L,the apoptosis rate was(17.4±1.7)%,G1(58.3±2.5)%,S(32.7±2.6)%,and M(6.3±1.4)%.EGCG promotes apoptosis.The higher the concentration of EGCG,the higher the ap-optosis rate,the higher the proportion of G1 cells,and the lower the proportion of s-phase cells.The higher the dose of EGCG,the higher the proportion of G1 cells and the higher the proportion of s-phase cells.Conclusion:EGCG can inhibit the proliferation,migration and apoptosis of HeLa cells cultured in vitro.
