血浆外泌体源性lncRNAH19和lncRNAMALAT1与膀胱癌的相关性研究 被引量:1

摘要 目的:探讨外泌体中lncRNAH19,MALAT1在膀胱癌中的表达水平及其诊断价值。方法:收集首次确诊且未接受手术、放化疗等任何处理的膀胱癌患者50例,选取同期在本院的健康体检者50例。分离并提取膀胱癌患者及正常体检者中的血浆外泌体,运用实时荧光定量(qRT-PCR)检测其血浆外泌体中lncRNAH19、MALAT1的表达情况,分析膀胱癌血浆外泌体中lncRNAH19、MALAT1的表达水平与临床病理参数的相关性,绘制ROC受试者工作曲线评价血浆外泌体中lncRNAH19、MALAT1对于膀胱癌的诊断价值。结果:膀胱癌患者组中lncRNAH19、MALAT1的表达水平显著高于健康对照组(P<0.05);膀胱癌外泌体中lncRNAH19、MALAT1的表达水平与年龄,性别,临床病理分级,临床病理分级,有无转移均无相关性。ROC受试者工作曲线lncRNAH19、MALAT1的曲线下面积(AUC)分别为0.971,0.878,lncRNAH19特异性为72%,敏感度为98%;lncRNAMALAT1特异性为84%,敏感度为92%。结论:血浆外泌体中lncRNAH19和lncRNAMALAT1的联合诊断可以提高早期膀胱癌诊断的特异性,具有较高的诊断价值,可作为早期膀胱癌筛查的一项比较理想的指标。 Objective:To investigate the expression levels of long non-coding RNA H19(lncRNA H19,H19)and long non-coding RNAs(lncRNAs)MALAT1 in exosomes in bladder cancer and their diagnostic value in bladder cancer.Methods:A total of 50 patients with bladder cancer who were diagnosed for the first time and did not undergo any treatment such as surgery or chemoradiotherapy were collected as the bladder cancer group,and 50 patients who underwent physical examination at the same time were selected as the healthy control group.Plasma exosomes of patients with bladder cancer and normal physical examination were isolated and extracted.The expressions of lncRNAH19 and MALAT1 in plasma exosomes were detected by real-time fluorescence quantifica-tion(qRT-PCR).The correlation between the expression levels of lncRNAH19 and MALAT1 in plasma exosomes of patients with bladder cancer and the clinopathological parameters was analyzed,and the receiver operating char-acteristic(ROC)curve was drawn to evaluate the diagnostic value of lncRNAH19 and MALAT1 in plasma exo-somes for bladder cancer.Results:The expression levels of the plasma exosomes lncRNAH19 and MALAT1 in the bladder cancer group were significantly higher than those in the healthy control group(P<0.05);There was no correlation between its expression level and age,sex,clinicopathological grade or metastasis.The area under ROC curve(AUC)of lncRNAH19 and MALAT1 were 0.971 and 0.878,respectively.LncRNAH19 had a specificity of 72%and a sensitivity of 98%.The specificity of lncRNAMALAT1 is 84%and the sensitivity is 92%.Conclusion:the combined diagnosis of lncRNAH19 and lncRNAMALAT1 in plasma exosomes can improve the specificity of ear-ly bladder cancer diagnosis,which is of high diagnostic value and can be used as an ideal indicator for early blad-der cancer screening.
