传统发酵樱菜中降解亚硝酸盐肠膜明串珠菌的分离鉴定及特性分析 被引量:5

摘要 从具有民族特色的延边朝鲜族地区采集24份传统发酵樱菜,从中分离筛选具有降解亚硝酸盐功能的菌株,通过形态观察、生理生化试验和16S rRNA序列分析进行鉴定,并对其生长特性、产酸特性、耐盐及耐酸碱特性进行研究。结果表明,从24份传统发酵樱菜中分离鉴定出1株具有降解亚硝酸盐功能的肠膜明串珠菌(Leuconostoc mesenteroides)JNZB003,菌株JNZB003对150μg/mL亚硝酸盐24 h时的降解率为95.92%,其最适生长温度为30℃,最适生长pH值为6.0,培养14 h生长进入稳定期,培养12 h pH值降至4.60,当NaCl含量为8%和pH值为3.0~10.0时能够生长,具有良好的降解亚硝酸盐能力、生长特性、产酸能力,且耐盐和耐酸碱。 A total of 24 traditional fermented Lepidium sativum samples were collected from Yanbian Korean area with national characteristics,and the strain with nitrite degradation capacity was isolated,screened,and identified by morphological observation,physiological and biochemical tests and 16S rRNA sequence analysis.At the same time,the growth characteristic,acid producing characteristic,salt,acid and alkali tolerance of the strain were studied.The results showed that the L.mesenteroides JNZB003 with nitrite degradation capacity was isolated and identified from 24 traditional fermented L.sativum samples,and the degradation rate of the strain JNZB003 to nitrite(150μg/ml)was 95.92%at 24 h.The optimum growth temperature and pH value of the strain JNZB003 was 30℃and 6.0,respectively.The strain JNZB003 grew into stable phase after culture 14 h,and the pH value decreased to 4.60 after culture 12 h.Under the conditions of NaCl 8%and pH value 3.0-10.0,the strain JNZB003 showed excellent nitrite degradation capacity,growth characteristics,and acid production capacity,as well as salt,acid and alkali tolerance.
