摘要
为了研究鸽源禽Ⅰ型副黏病毒辽宁分离株的遗传变异情况及分子生物学特征。根据GenBank公布的鸽源禽Ⅰ型副黏病毒Pi/SH/CH/0168/2013毒株设计16对引物,对辽宁分离株全基因分段进行扩增并测序,将测序结果依次拼接得到病毒的全基因组,然后进行序列的比对分析。结果表明:辽宁分离株基因全长15192 nt,并命名为LNPPMV17。F0裂解位点的氨基酸序列为112R-R-Q-K-R-F117,结合F蛋白全基因遗传进化分析,辽宁分离株属于ClassⅡ基因Ⅵ型;并发现鸽副黏病毒M蛋白第36和68位点氨基酸与其他禽类副黏病毒有明显不同,这种差异可以作为区别其他禽类Ⅰ型副黏病毒的标志;全基因序列分析结果表明国内鸽源禽Ⅰ型副黏病毒的同源性较高,这可能是近年来赛鸽业迅速发展,鸽子流通增加,交叉感染所致。
The research was performed to study the genetic variation and molecular biological characteristics of pigeon-sourced paramyxovirus type 1 isolated in Liaoning Province.16 pairs of oligonucleotide primers were designed,according to the Pi/SH/CH/0168/2013 strain of GenBank,and were used to amplify the genome of the virus in a sectional manner and then sequenced it.The sequenced results were spliced into the whole genome of the strain,and alignment was performed.The results showed that the whole genome of the Liaoning strain was 15192 nt long and was named LNPPMV17;its amino acid sequence of F0 cleavage site was 112R-R-Q-K-R-F117.Combined with complete fusion gene phylogenetic analysis,the Liaoning strain was classified as ClassⅡgenotypeⅥavian paramyxovirus.It was found that the amino acids at the 36 and 68 loci of the matrix protein of pigeon-sourced paramyxovirus type 1 were significantly different from those of the other avian paramyxoviruses,and two acid differences might be a marker to distinguish other avian paramyxoviruses type 1.The whole-genome sequence analysis showed that the homology of pigeon-sourced avian paramyxovirus type 1 was high in China,which might have been caused by the rapid development of pigeon racing sports in recent years,the increased mobility of the birds,and the accompanied high cross-infection with viruses.
