发酵对蛋白桑1-脱氧野尻霉素提取量的影响 被引量:5

摘要 本试验旨在通过研究发酵及提取方法对蛋白桑中1-脱氧野尻霉素(DNJ)提取量的影响,确定DNJ提取合理的前处理方法。试验用酵母菌和枯草芽孢杆菌的混合菌液对蛋白桑在不同条件下进行发酵,筛选出最佳发酵样品后,进行干燥处理。分别用水提法、酸提法和醇提法对所筛选样品中的DNJ进行提取,采用高效液相色谱法测定DNJ含量。结果表明:三种方法对最佳发酵条件下(接种量6%,发酵温度35℃,发酵时间96 h)的发酵样品DNJ提取量均为最高。对不同的样品进行提取时,醇提法提取效果普遍优于酸提法和水提法,发酵样品DNJ提取量比对照组高,最高可达34.07%,差异显著(P≤0.05)。结果说明发酵可以提高DNJ的提取量,在DNJ提取方法选择上,醇提法效果最佳。 To study the effect of different fermentation conditions and extraction methods on the extraction of 1-Deoxyrummmycin(DNJ)in the mulberry.Mulberry samples were fermented by yeast and Bacillus subtilis mixed bacteria broth under different conditions.Then the best samples were selected and dried.The samples of DNJ were extracted by water extraction,acid extraction and alcohol extraction and determined by high performance liquid chromatography.The results showed that the amount of DNJ from the fermented sample under the optimum fermentation conditions(inoculation 6%,fermentation temperature 35℃,fermented for 96 h)was the highest when extracted in three methods.When extracting different samples,the alcohol extraction method was generally better than acid extraction and water extraction.The DNJ amount of sample extracted by ethanol extraction method was 34.07%higher than that of the control group(P<0.05).This result indicated that fermentation could improve the extraction amount of DNJ and the best extraction method was alcohol extraction.
