摘要
目的观察化聚消积方对肥胖伴高血压大鼠早期肾损伤的影响,探讨其作用机制。方法 40只SD大鼠随机分为正常组10只和高盐高脂组30只,高盐高脂组给予高盐高脂饮食干预2周,分为模型组、阳性药组及中药组,每组10只。药物干预6周,测量大鼠血压,记录体质量,ELISA检测尿微量白蛋白(MAU),生化法检测血肌酐;肾脏行HE及PAS染色观察组织病理学形态;放免法检测血浆血管紧张素Ⅱ(AngⅡ)、血管紧张素转换酶(ACE)水平;RT-PCR检测肾脏肾素(Renin)、AngⅡ、AngⅡ1型受体(AT1R)的mRNA表达,免疫组化检测肾小管局部AT1R、AT2R蛋白表达。结果高盐高脂饮食干预后大鼠血压明显升高、体质量明显增加,与正常组比较,模型组大鼠MAU水平明显升高;与模型组比较,阳性药组和中药组大鼠MAU水平明显降低,血浆AngⅡ及ACE水平未见明显变化,肾脏局部Renin、AngⅡ及AT1R基因表达明显降低,肾小管AT1R及AT2R蛋白表达明显降低。结论化聚消积方可能通过抑制肾脏及肾小管局部肾素-血管紧张素系统水平而改善肥胖伴高血压大鼠肾脏损伤的作用。
Objective To observe the effects of Huaju Xiaoji Prescription on early renal injury in obese rats with hypertension;To explore its mechanism.Methods Totally 40 SD rats were divided into normal group(10 rats)and high salt and high fat group(30 rats).The high salt and high fat group was divided into model group,positive medicine group and TCM group after 2 weeks of high salt and high fat diet intervention.After 6 weeks of drug intervention,blood pressure was measured;body weight was recorded;microalbumin uria(MAU)was detected by ELISA;creatinine in blood was detected by biochemical method.Histopathological morphology was observed by HE and PAS staining.Plasma levels of AngⅡand ACE were detected by radioimmunoassay.Renin,AngⅡand ACE mRNA expression in kidney were detected by RT-PCR.The protein expressions of AT1R and AT2R in renal tubules were detected by immunohistochemistry.Results After the intervention with high salt and high fat diet,the blood pressure and body weight of rats significantly increased.Compared with the normal group,the MAU of rats in the model group significantly increased.Compared with the model group,the MAU in both positive medicine and TCM group decreased significantly,while the level of plasma AngⅡand ACE did not change.The mRNA expressions of Renin,AngⅡand AT1R in the kidney,and the expressions of AT1R and AT2R protein in the renal tubules in both treatment groups decreased significantly.Conclusion Huaju Xiaoji Prescription may improve the renal injury of obese rats with hypertension by inhibiting the level of rein-angiotension system in kidney and renal tubules.
