基于生物信息学的疏肝利胆和胃方组方特点及治疗反流性食管炎机制分析

Analysis on Prescription Characteristics of Shugan Lidan Hewei Prescription and Its Mechanisms for Reflux Esophagitis Based on Bioinformatics

目的基于生物信息学方法研究疏肝利胆和胃方的组方特点及治疗反流性食管炎的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)收集疏肝利胆和胃方中10味中药的活性成分及其作用靶点,进行主坐标分析(PCoA),研究其组方特点;构建化合物-靶点网络及蛋白相互作用网络,并依托CTD数据库形成反流性食管炎背景网络,进行靶点映射,分析疏肝利胆和胃方治疗反流性食管炎机制。结果化合物-靶点网络包含212个有效成分及862个分子靶点。方内协同增效的药对包括黄芩-枳实和白芷-砂仁。半夏为该方的核心药物,该方治疗反流性食管炎的核心靶点为PTGS2和BCL2L1,涉及NF-κB信号通路、IL-17信号通路、TNF信号通路等。结论本研究初步验证了疏肝利胆和胃方的组方特点,并揭示该方治疗反流性食管炎的主要有效成分起效机制,可为进一步探讨其作用机制奠定基础。

Objective To study the prescription characteristics and mechanism of Shugan Lidan Hewei Prescription for reflux esophagitis(RE)based on bioinformatics.Methods The active components and action targets of 10 kinds of Chinese materia medica in the prescription were collected by TCMSP and analyzed by principal component analysis(PCoA),and prescription characteristics were studied.Compound-target network and protein-protein interaction network were constructed,and the background network of RE was formed based on CTD database.Target mapping was carried out.Mechanism of Shugan Lidan Hewei Prescription for RE was analyzed.Results Compound-target network consisted of 212 active components and 862 molecular targets.The synergistic medicinal pairs included Scutellariae Radix-Aurantii Fructus Immaturus and Angelicae Dahuricae Radix-Amoni Fructus.Pinelliae Rhizoma was the core medicine of this prescription.The core targets of the prescription for treating RE were PTGS2 and BCL2L1.The pathways involved were NF-κB,IL-17,TNF signaling pathway,etc.Conclusion The results of this study preliminarily validate the characteristics of Shugan Lidan Hewei Prescription,and reveal the effective mechanism of its main active components for the treatment of RE,which lays a good foundation for further exploring the mechanism of Shugan Lidan Hewei Prescription.
