百合水溶液对麻黄素肺组织氧化损伤和Caspase-3,c-Fos表达的影响 被引量:1

摘要 为了探讨百合对麻黄素肺组织氧化损伤和Caspase-3,c-Fos表达的影响,选90只昆明小鼠分为对照组、麻黄素组和百合组,采用递增剂量给小鼠连续(1~5, 5~10, 11~15 d)腹腔注射(2.0,4.0,6.0 g·L-1)麻黄素15 d(每天2次,每次0.2m L),注射麻黄素1 h后,再给小鼠灌胃百合水溶液0.2 m L,使用称量法检测小鼠的体重和肺重,利用比色法检测小鼠肺组织丙二醛(MDA)含量的变化,用免疫组织化学法检测肺组织Caspase-3,c-Fos蛋白表达的变化.结果表明,与对照组相比,麻黄素组小鼠体重降低,肺指数升高,肺组织MDA含量升高,Caspase-3和c-Fos蛋白的阳性表达增强(P<0.05或P<0.01);而百合组与麻黄素组相比,小鼠体重升高,肺指数降低(P<0.05),肺组织MDA的含量明显降低(P<0.05或P<0.01),肺组织Caspase-3和c-Fos蛋白阳性表达的平均光密度值减小(P<0.05或P<0.01).说明百合水溶液能提高肺组织细胞的抗氧化能力,抑制肺组织Caspase-3,c-Fos蛋白的阳性表达,对肺组织有一定的保护作用. This paper was explored the effects of Lilium soup on ephedrine lung histological oxidative damage and specific protein expression.90 Kunming mice were randomly divided into control group,ephedrine group and Lilium group.The mice were intraperitoneally injected escalation with doses(2.0,4.0,6.0 g·L-1)ephedrine 0.2 mL continuous(1~5,6~10 and 11~15 d)for 15(twice a day).After injection of ephedrine for 1 h,Lilium group was intragastric administration with 0.2 mL Lilium soup.The body weight and lung weight were recorded,and changes of lung exponential were counted,the changes of malondialdehyde(MDA)content in mice lung were analyzed by colorimetry,and the changes of Caspase-3,c-Fos expression in lung tissue were determined by immunohistochemical method.The results showed that the body weights of ephedrine group is significantly decreased,while the lung exponential is increased significantly.The MDA content in the lung of ephedrine group is significantly increased.The intensities of Caspase-3 and c-Fos protein positive expression in the lung is increased significantly(P<0.05 or P<0.01).However,the body weight of Lilium group is significantly increased but the lung exponential is significantly decreased(P<0.05),the MDA content in the lung of Lilium group is significantly decreased(P<0.05 for P<0.01),and the intensities of Caspase-3 and c-Fos protein positive expression in the lung are significantly decreased(P<0.05 or P<0.01).Lilium soup can increase antioxidance ability of lung tissue,restrain expression of Caspase-3 and c-Fos protein,and reduce lung tissue damage induced by ephedrine effectively.
