头腹针疗法对气虚质缺血性中风大鼠干预机制研究

Intervention Mechanism of Scalp and Abdominal Acupuncture on Stroke of Qi Deficiency in Rats

目的:分析头腹针疗法对气虚质中风大鼠的作用机制。方法:健康雄性Wistar大鼠40只,气虚质造模,成功后随机分为模型组、治疗组(头腹针疗法),每组15只模型大鼠,通过MCAO造模;另外10只为假手术组,不操作。3 d、7 d、14 d及21 d 4个时间点采用神经功能评分、行为学评估、PCR检测Hes1 mRNA、Western-Blot检测Hes1蛋白。结果:神经功能评分、行为学评估7 d、14 d评分治疗组优于模型组(P<0.05或P<0.01);PCR检测Hes1 mRNA阳性表达7 d、14 d治疗组优于模型组(P<0.05或P<0.01);Western-Blot检测Hes1蛋白14 d治疗组优于模型组(P<0.05)。结论:头腹针疗法对气虚质中风大鼠神经干细胞具有干预作用。

Objective:To analyze the mechanism of scalp and abdominal acupuncture on stroke of qi deficiency in rats.Methods:40 healthy male Wistar rats were selected to establish qi deficiency model by MCAO.The successful modeling were randomly divided into the model group(n=15),the treatment group(n=15)and the sham-operation group(n=10).The treatment group was treated with scalp and abdominal acupuncture.Neurological scores and behavioral evaluation were assessed,Hes1 mRNA and Hes1 protein were detected by PCR and Western Blot method respectively on 3rd,7th,14th and 21st day.Results:The neurological scores and behavioral evaluation were better in the treatment group that those in the model group on 7th day and 14th day(P<0.05 or P<0.01).The positive expression of Hes1 mRNA was more significant in the treatment group than that in the model group on 7th day and 14th day(P<0.05 or P<0.01).The expression of Hes1 protein was better in the treatment group than that in the model group on 14th day(P<0.05).Conclusion:Scalp and abdominal acupuncture had obvious intervention effect on neural stem cells in rats with stroke of qi deficiency.
