摘要
目的:观察电针神门、四神聪对卵巢摘除雌性大鼠睡眠时相的影响,并通过其对脑脊液(CSF)中前列腺素D2(PGD2)系统的影响初步探讨其作用机制。方法:30只清洁级健康Wistar雌性大鼠随机分为3组,即假手术组、模型组与电针组,每组10只。手术摘除雌性大鼠双侧卵巢联合电刺激复制大鼠围绝经期失眠模型。同时采用电针神门、四神聪的方法连续治疗21 d后,监测各组大鼠12 h肌电图、脑电图,分析睡眠时相。酶联免疫法测定脑脊液中L-PGDS、PGD2、PGE2水平,免疫组化法测定下丘脑腹外侧视前(VLPO)区γ-氨基丁酸(GABA)神经递质水平。结果:与假手术组比较,模型组大鼠浅睡眠(LS)、慢波睡眠(SWS)、快动眼睡眠(REMS)与睡眠总时长(TS)均明显缩短(P<0.05),各睡眠时相所占比例发生改变,CSF中L-PGDS、PGD2水平明显降低,PGE2显著升高,VLPO中GABA神经递质水平显著下降(P<0.05)。与模型组比较,电针组大鼠SWS、REMS显著延长(P<0.05),REMS期睡眠所占比例显著恢复(P<0.05),CSF中L-PGDS、PGD2水平以及VLPO中GABA神经递质水平显著升高(P<0.05),LS、TS以及CSF中PGE2水平变化差异无统计学意义(P>0.05)。结论:电针神门、四神聪可以改善卵巢摘除大鼠的睡眠质量,其作用机制与调节CSF中PGD2系统平衡以及VLPO中GABA神经递质的水平密切相关。
Objective:To investigate the effects of electro-needling Shenmen(HT7)and Sishencong(EX-HN1)on sleep phases of ovariectomized rats and to explore its mechanism via prostaglandin D2(PGD2)system of cerebrospinal fluid(CSF).Methods:30 healthy Wistar female rats were randomly divided into the sham-operated group,the model group and the electro-acupuncture group,with 10 rats in each group.Perimenopausal insomnia model was established by ovariectomy combined with electrical stimulation.The rats in electro-acupuncture group were treated by electro-needling HT7 and EX-HN1 for 21 days continuously.EMG and EEG were monitored for 12 hours to analyze sleep phases.The levels of L-PGD synthetase(L-PGDS),PGD2 and PGE2 in CSF,as well as GABA in ventrolateral preoptic(VLPO)nucleus were evaluated by Elisa method and immunohistochemistry respectively.Results:Compared to the sham-operated group,the duration of light sleep(LS),slow wave sleep(SWS),rapid eye movement sleep(REMS)and total sleep(TS)shortened obviously in the model group(P<0.05);the proportion of sleep phases changed;the levels of L-PGDS,PGD2 in CSF and GABA in VLPO decreased markedly(P<0.05),whereas the level of PGE2 in CSF increased dramatically(P<0.05).Compared to the model group,the duration of SWS and REMS prolonged significantly,the proportion of REMS improved obviously(P<0.05);the levels of L-PGDS,PGD2 and GABA increased significantly in the electro-acupuncture group(P<0.05);there were no statistical differences in the changes of LS,TS and PGE2 in the electro-acupuncture group(P>0.05).Conclusion:Electro-needling HT7 and EX-HN1 can improve sleep quality of ovariectomized rats.Its mechanism is related to rebalancing PGD2 system of CSF and regulating the level of GABA in VLPO.
