摘要
目的:探讨利湿化瘀中药对慢性盆腔炎大鼠输卵管组织Fas/FasL通路的影响。方法:沿大鼠子宫交叉处向卵巢方向、双侧子宫内分别注入25%苯酚胶浆0.04 mL,建立慢性盆腔炎大鼠模型。造模8 d后,随机分成模型组、中药高剂量组(8 g/kg)、中药低剂量组(4 g/kg)和阳性对照妇乐组(2.5 g/kg),各组分别灌胃给药21 d,另设正常对照组10只。采用Western Blot法检测输卵管组织Caspase-3、Casepase-8、FasL蛋白表达;原位杂交法检测输卵管Caspase-8mRNA表达。结果:与模型组比较,中药高剂量组、中药低剂量组、妇乐组Caspase-3、Casepase-8、FasL蛋白表达均明显降低(P<0.05),中药高剂量组效果明显于中药低剂量组中、妇乐组;模型组可见少量Caspase-8mRNA表达,中药低剂量组偶见,中药高剂量组及妇乐组均未见其表达。结论:利湿化瘀中药可通过降低Caspase-3、Casepase-8、FasL蛋白及Caspase-8mRNA的异常表达、延缓细胞凋亡、改善盆腔炎输组织粘连及纤维化。
Objective:To explore the effects of Removing Dampness and Blood Stasis Chinese medicine on the Fas/FasL pathway of fallopian tubal tissue in rats with chronic pelvic inflammatory disease.Methods:the rat model of chronic pelvic inflammatory disease was established by injecting 0.24 mL of 25% phenolic mucilage into the ovary and bilateral uterus along the uterus.After 8 days of modeling,40 rats were randomly divided into model group,Chinese medicine high-dose group(8 g/kg)group,Chinese medicine low-dose group(4 g/kg)group and positive control Fule Granule group(2.5 g/kg),and oral administration respectively for 21 days,10 healthy rats were taken as normal group.The expressions of Caspase-3,Casepase-8 and FasL protein of rats’fallopian tubal tissue were detected by Western Blot,the expression of Caspase-8 mRNA was detected by in situ hybridization.Results:Compared with the model group,the expressions of Caspase-3,Casepase-8 and FasL protein of fallopian tubal tissue in the Chinese medicine high-dose group,Chinese medicine low-dose group and Fule Granule group were significantly lowered(P<0.05),the effects of Chinese medicine high-dose group were better than those of Chinese medicine low-dose group group and the Fule Granule group;a small amount of Caspase-8mRNA expression was seen in the model group,occasional-seen in the Chinese medicine low-dose group and no-seen in the Chinese medicine high-dose group groups and the Fule Granule group.Conclusion:Removing Dampness and Blood Stasis Chinese Medicine can effectively treat chronic pelvic inflammatory disease by reducing the abnormal expression of Caspase-3,Casepase-8,FasL protein and Caspase-8 mRNA,delaying cell apoptosis,improving tissue adhesion and fibrosis.
