五福饮对椎间盘退变大鼠模型血清及椎间盘炎性因子表达的影响

Effects of Wufuyin(五福饮)on Inflammatory Factors Expressions in Serum and Intervertebral Disc of Rats with Degenerative Disc

目的:观察五福饮对椎间盘退变大鼠模型血清及椎间盘炎性因子TNF-α(肿瘤坏死因子)、IL-1(白细胞介素-1)、IL-6(白细胞介素-6)、MMP-3(金属基质蛋白酶-3)的影响。方法:将48只雄性SD大鼠随机分成3组,每组各16只,即正常对照组、退变模型组及五福饮干预组;应用细针穿刺纤维环结合肿瘤坏死因子-α椎间盘内注射的方法构建椎间盘退变大鼠模型,造模成功后,3组大鼠分别给予等量的生理盐水、生理盐水及五福饮灌胃治疗8周。分别摘除各组大鼠目标椎间盘,HE染色观察病理变化,采用ELISA法测定各组大鼠血清及椎间盘TNF-α、IL-1、IL-6、MMP-3的含量。结果:病例观察可见五福饮干预组和退变模型组的目标椎间盘均有不同程度的退变,五福饮干预组椎间盘退变程度相对较轻。退变模型组大鼠血清及椎间盘内的TNF-α、IL-1、IL-6、MMP-3与正常对照组相比明显升高(P<0.05);五福饮干预组与退变模型组相比血清及椎间盘内的TNF-α、IL-1、IL-6、MMP-3显著降低(P<0.05)。结论:五福饮可通过降低TNF-α、IL-1、IL-6的表达抑制MMP-3的表达,延缓椎间盘内基质的降解,防治椎间盘退变。

Objective:To observe the effects of Wufuyin on the expressions of inflammatory factors(TNF-α,IL-1,IL-6,MMP-3)in intervertebral disc and serum of rats with degenerative disc.Methods:48 SD male rats were randomly divided into control group,degeneration model group and Wufuyin intervention group,16 cases in each group.Models of intervertebral disc degeneration were used fine needle fibrous ring puncture combined with TNF-α intradiscal injection to make,groups were given normal saline,saline and Wufuyin gavage for 8 weeks respectively,the characteristics of intervertebral disc degeneration were observed by HE staining,the levels of TNF-α,IL-1,IL-6 and MMP-3 in the serum and intervertebral discs of rats in each group were determined by ELISA.Results:The pathological examination showed that different degrees of degeneration were found in Wufuyin intervention group and the degeneration model group,but the degree of degeneration of intervertebral disc was relatively mild in the Wufuyin intervention group.There were significant differences in the contents of TNF-α,IL-1,IL-6 and MMP-3 in serum and intervertebral disc among the three groups(P<0.05),the levels of TNF-α,IL-1,IL-6 and MMP-3 in serum and intervertebral disc in the degenerative model group were significantly higher than those in the normal control group(P<0.05),compared with the degenerative model group,the contents of TNF-α,IL-1,IL-6 and MMP-3 in serum and intervertebral disc in the Wufuyin intervention group were significantly decreased,with significant difference(P<0.05).Conclusion:Wufuyin can prevent and cure intervertebral disc degeneration,and the mechanism may be related to delay the degradation of intervertebral disc matrix by reducing the levels of TNF-α,IL-1,IL-6 to inhibit MMP-3 expression.
