豆豉对小鼠雄激素源性脱发的改善作用及机制研究 被引量:7

摘要 目的:探讨豆豉对雄激素源性脱发小鼠的改善作用及作用机制。方法:将72只小鼠随机分为正常组、模型组、阳性对照组和豆豉高、中、低剂量组6组,每组12只。除正常组外,其余各组小鼠皮下注射丙酸睾酮溶液建立AGA模型,阳性药组灌胃非那雄胺,豆豉各剂量组给予豆豉溶液3.25、6.50、13.00 g/(kg·d)灌胃,正常组和模型组灌胃等体积蒸馏水,观察小鼠一般状况,称量脱发重量评价脱发程度,光镜观察毛囊组织病理学改变,ELISA检测血清性激素水平,RT-PCR方法对皮肤组织TGF-β2(转化生长因子-β2)、Caspase-3mRNA(凋亡因子)含量进行测定。结果:造模5周后,小鼠背部毛色失去光泽并呈灰暗颜色,于7周左右逐渐出现毛发脱落。各给药组小鼠脱毛较模型组明显减轻;病理学观察可见与正常组相比,模型组小鼠毛囊总数明显变少,密度减小,毳毛明显增多,各给药组终毛多,毳毛较少。血清睾酮(T)、雌二醇(E 2)测定结果显示模型组T含量明显高于正常组,差异非常显著(P<0.01),各给药组T含量均低于模型组,具有显著性差异(P<0.05);各给药组E 2含量均高于模型组,有明显差异(P<0.05)。与正常组比较,模型组小鼠皮肤组织TGF-β2、Caspase-3mRNA表达明显增加,各给药组小鼠皮肤组织TGF-β2、Caspase-3mRNA较模型组明显降低。结论:豆豉对AGA小鼠的作用机制可能是通过调节小鼠体内性激素水平,抑制T转化为二氢睾酮(DHT),削弱二氢睾酮诱导的TGF-β信号通路,抑制毛囊细胞凋亡,从而达到防脱发的作用。 Objective:To explore the improvement effect and mechanism of Soybean Meal on androgenetic alopecia(AGA)mice.Methods:72 mice were randomly divided into normal group,model group,positive control group and Soybean Meal high,medium and low dose groups,12 in each group.Except for the normal group,mice in other groups were injected subcutaneously with testosterone propionate solution to establish the AGA model.Each dose group of Soybean Meal was given Soybean Meal solution 3.25,6.50,13.00 g/(kg·d),and the normal group and model group were gavaged with equal volume of distilled water.Observing the general condition of mice,weighing the weight of hair loss,evaluating the degree of hair loss,observing the pathological changes of hair follicles under light microscope,detecting the level of serum sex hormones by ELISA,TGF-β2(transforming growth factor-β2),Caspase-3 mRNA(apoptosis factor)content in skin tissue were measured by RT-PCR method.Results:After 5 weeks of modeling,the hair on the back of the mice tarnished and showed a dull color,and hair loss gradually appeared around 7 weeks.Compared with the model group,the hair loss of mice in each medication administration group was significantly reduced;pathological observation showed that the total number of hair follicles in the model group was significantly reduced,the density was reduced,and the vellus hair was significantly increased compared with the normal group,less hairy,T content of the model group was significantly higher than that of the normal group(P<0.01);E2 content of each medication administration group was higher than that of the model group(P<0.05).Compared with the normal group,the expression of TGF-β2 and Caspase-3 mRNA in the skin tissue of the model group was significantly increased,and the expression of TGF-β2 and Caspase-3 mRNA in the skin tissue of each medication administration group was significantly lower than those of the model group.Conclusion:The mechanism of Soybean Meal on AGA mice may be by regulating sex hormone levels in mice,inhibiting the conversion of T into dihydrotestosterone(DHT),weakening the dihydrotestosterone-induced TGF-β signaling pathway,inhibiting hair follicle cell apoptosis,and thus preventing The role of hair loss.
