摘要
目的探索丝素蛋白(SF)/壳聚糖(CS)/羟基磷灰石(nHA)生物膜的生物相容性及促成骨性能。方法以冷沉淀法制备2组复合膜,将SF/CS/nHA膜作为实验组,SF/CS膜作为对照组,并以载玻片作为空白组;将MG-63细胞接种于以上3组,计算细胞的黏附率及增殖率,并通过分析MG-63分泌的碱性磷酸酶活性来研究复合膜的促成骨性能。结果 MG-63细胞在培养的第1、4、7小时,实验组的细胞黏附率吸光值为0.316±0.007、0.460±0.007和0.519±0.006,均高于对照组及空白组(P<0.05);MG-63细胞在培养的第1、3、5天,实验组的细胞增殖率吸光值为0.342±0.009、0.612±0.013和0.676±0.009,实验组在第3天的增殖情况高于对照组(P<0.05);MG-63细胞在培养的第7、14、21天,实验组的细胞碱性磷酸酶吸光值为0.814±0.006、1.046±0.071和1.222±0.012,均高于对照组及空白组(P<0.05)。结论 SF/CS/nHA具有良好的生物相容性及具有一定的骨引导性,能够诱导成骨细胞的粘附,增殖及促进其成骨。
Objective To explore the biocompatibility and osteoporosis of the Silk fibroin(SF)/Chitosan(CS)/hydroxyapatite(nHA) membrane. Methods SF/CS/nHA membrane were prepared by cryoprecipitation method. The adhesion rate,proliferation rate and alkaline phosphatase of MG-63 cell in the SF/CS/nHA(experimental group), SF/CS(control group) and galass slides(blank contril group) were Observed. Results The cell anhesion optical density of experimental group was higher than control group and blank contril group in 1 h(0.316 ± 0.007), 4 h(0.460 ± 0.007) and 7 h(0.519 ± 0.006)(P<0.05). The cell proliferation optical density of experimental group was higher than that of control group in 3 d(0.612 ± 0.013)(P<0.05). The alkaline phosphatase optical density of experimental group was higher than that of control group and blank control group in 7 d(0.814±0.006), 14 d(1.046±0.071) and 21 d(1.222±0.012)(P<0.05). Conclusion SF/CS/n HA membrane have good biocompatibility and osteoporosis.
