获得性免疫缺陷综合征患者易感抑郁症差异表达基因的生物信息学分析

摘要 目的探索获得性免疫缺陷综合征(AIDS)患者易感抑郁症的关键差异表达基因,为进一步研究寻找靶点。方法通过GEO数据库下载AIDS合并抑郁症患者和AIDS患者额叶皮层灰质细胞的基因芯片数据集GSE17440,利用R语言挖掘差异表达基因,通过STRING、metascape等工具进行生物信息学分析。结果挖掘出差异表达基因71个,其中上调基因16个,下调基因55个。经过不同的富集工具富集,差异表达基因主要集中于细胞凋亡通路。构建蛋白与蛋白互相作用的网络图显示,差异表达基因的表达产物大部分存在密切的相互作用,AIDS合并抑郁症患者脑部组织免疫防御反应异常激活。结论通过对AIDS合并抑郁症患者和AIDS患者差异表达基因的分析,对其差异基因有了系统的认识,针对关键基因的大样本研究及网络化基因分析是未来的研究方向。 Objective To explore key differentially expressed genes in patients with acquired immunodeficiency syndrome(AIDS)susceptible to depressive disorder,and find targets for further research.Methods The gene chip data set GSE17440 of frontal cortex gray matter cells of AIDS patients with depressive disorder and AIDS patients was downloaded through the GEO database,and the differentially expressed genes were mined by R language,and bioinformatics analysis was performed via tools such as STRING and metascape.Results A total of 71 differentially expressed genes were identified,including 16 up-regulated genes and 55 down-regulated genes.After enrichment with different ways,the results showed that the differentially expressed genes were mainly concentrated in the apoptotic pathway.The network map of protein-protein interaction of translatable proteins was constructed and showed that most of the expression products of these genes had close interactions,and the pathway of defense response to other organism were abnormally activated in brain tissue of AIDS patients with MDD.Conclusion The systematic understanding of the differential genes through the analysis of differentially expressed genes between AIDS patients with MDD and AIDS patients.Large sample research on key genes and network genetic analysis are future research directions.
