Mex3c参与调控小鼠下丘脑Arc、VMH神经核团C-FOS表达的作用 被引量:4

摘要 目的探索秀丽隐杆线虫肌肉表达物3(Mex3c)诱导下丘脑神经核团C-FOS蛋白(原癌基因表达产物)表达的作用。方法 8周龄正常和Mex3c-/+小鼠各15只,随机分为正常对照组、正常瘦素组、正常饥饿激素组、Mex3c-/+对照组、Mex3c-/+瘦素组、Mex3c-/+饥饿激素组,每组5只,Mex3c-/+各组小鼠注射干预及数量同正常一样且均采取腹腔注射。注射2 h后麻醉灌注后取小鼠脑组织,免疫组织化学染色分析下丘脑弓状核(Arc)、腹内侧核(VMH)核团C-FOS蛋白的表达量。结果病理学染色正常小鼠和Mex3c-/+小鼠未见明显差异。免疫组化结果显示,正常对照组小鼠下丘脑Arc、VMH核团中C-FOS蛋白表达量较低,在注射瘦素和饥饿激素2 h后C-FOS的表达量均增加(P<0.05);Mex3c-/+对照小鼠相比正常正常对照小鼠,C-FOS为高表达(P<0.05),与正常瘦素组小鼠差异无统计学意义(P>0.05);给予Mex3c-/+小鼠饥饿激素干预后,C-FOS的表达量下调(P<0.05),正常瘦素组和饥饿激素组与Mex3c-/+对照组和瘦素组之间C-FOS表达量差异均无统计学意义(P均>0.05)。结论Mex3c可以诱导小鼠下丘脑Arc、VMH核团C-FOS的表达进而参与神经信号转导。 Objective To explore the effect of muscle expression 3(Mex3c) of Caenorhabditis elegans on the expression of C-FOS protein(Proto-oncogene C-FOS expression product)in hypothala mic nucleus.Methods Totally 30 eight-week-old wild-type and Mex3c gene-deficient mice constructed of by CRISPR/Cas9 system were randomly divided into normal control group(n=5),normal leptin group(n=5),normal ghrelin group(n=5),Mex3c-/+control group(n=5),Mex3c-/+leptin group(n=5),Mex3c-/+ghrelin group(n=5). Mex3c-/+mice were injected intraperitoneally in the same amount as normal. The expression of C-FOS protein in hypothalamic arcuate nucleus(ARC)and ventromedial nucleus(VMH)was analyzed by immunohistochemistry. Results Pathological staining showed no significant difference between normal mice and Mex3c-/+mice. The results of immunohistochemistry showed that the expression of C-FOS protein in the hypothalamic arc and VMH nuclei of the normal control group was lower,and the expression of c-fos increased 2 hours after injection of leptin and ghrelin(P<0.05). The expression of C-FOS in the Mex3c-/+control group was higher than that in the normal control group(P<0.05),and there was no significant difference between the Mex3c and the normal leptin group(P>0.05). After intervention with ghrelin in Mex3c-/+mice,the expression of C-FOS was significantly reduced but still higher than the normal control group(P<0.05). There was no significant difference in C-FOS expression between the normal leptin,ghrelin group,Mex3c-/+normal saline contro and leptin group(P all>0.05). Conclusion Mex3c can induce the expression of C-FOS in the hypothalamic Arc and VMH nuclei in mice and participate in neural signal transduction.
