摘要
【目的】为了探究鸭FMO3基因型、动物油脂及其互作对其FMO3基因表达水平的影响.【方法】将79只鸭平均分为2组,用含10%动物油脂和不含动物油脂的2种饲料分别饲养,通过对鸭FMO3基因的第二、六、七外显子的SNPs进行检测,qPCR检测鸭肝、肾中FMO3基因的表达水平,并对FMO3基因SNPs和动物油脂与FMO3基因在肝、肾中的表达进行关联分析.【结果】在鸭FMO3基因SNPs检测中发现cDNA第12bp A/G碱基突变位于FMO3蛋白结构的FAD结合区,与FMO3在肝脏中的表达水平相关,命名为V12I错义突变.GG突变纯合子个体在饲喂添加10%动物油脂时,肝脏中FMO3基因的表达量显著低于饲喂不含动物油脂的处理组(P<0.05),AA、AG2种基因型在是否饲喂含10%的动物油脂2个处理组中肝脏FMO3基因的表达量均没有显著性差异(P>0.05).而是否添加10%的动物油脂、FMO3基因型以及二者的交互作用对肾脏FMO3基因的表达没有显著性影响(P>0.05).【结论】综上表明,饲料中添加10%动物油脂可抑制肝脏FMO3基因的表达,且对GG基因型的影响显著.
【Objective】In order to explore the effects of FMO3 genotype,animal fats and their interactions on FMO3 gene expression levels.【Method】79 ducks were divided into 2 groups,which were fed with 2 diets with different animal fat contents.The SNPs of the second,sixth,and seventh exons of duck FMO3 gene were detected,qPCR was used to detect the expression of FMO3 gene in duck liver and kidney.The association between FMO3 gene SNPs,animal fats and FMO3 gene expression in liver and kidney was studied.【Result】12th bp A/G base mutation of cDNA was found in FAD binding region of FMO3 protein structure,which associated with the expression of FMO3 in the liver and was named as V12I missense mutation.The genotype had no significant effect on the expression of the liver FMO3 gene.When the GG mutant homozygous individuals were fed with 10%animal fat,the expression of FMO3 gene in liver was significantly lower than another treatment group(P<0.05).There was no significant difference in the expression of FMO3 gene between AA genotypes and AG genotypes in two groups(P>0.05).Animal fat,genotype and their interaction had no significant effect on the expression of FMO3 in kidney(P>0.05).【Conclusion】The addition of 10%animal fat to diet inhibits the expression of FMO3 gene in liver and shows a significant effect on GG genotype.
