长江上游中华纹胸鮡遗传多样性及遗传结构研究 被引量:2

Genetic diversity and structure of Glyptothorax sinensis in upper Yangtze River

摘要 为研究中华纹胸鮡(Glyptothorax sinensis)遗传多样性和遗传结构,对长江上游9个中华纹胸鮡群体(巴南、江津、合江、泸州、南溪、犍为、巧家、宁南和攀枝花)的Cyt b基因和COI基因进行分析。结果显示:341条Cyt b序列共检测出80个变异位点,65个单倍型,平均单倍型多样性(Hd)和平均核苷酸多样性(Pi)分别为0.845和0.007。109条COI序列共检测出31个变异位点,23个单倍型,Hd指数和Pi指数分别为0.774和0.005。分子方差分析(AMOVA)显示FST>0.5,说明中华纹胸鮡采样点群体间出现明显分化。构建的NJ树和单倍型网络结构图显示单倍型聚类形成三个高支持率的分支。 In this study,the mitochondrial DNA cytochrome b(Cyt b)and cytochrome C oxidase subunitⅠ(COI)sequences were used to analyse population genetics of Glyptothorax sinensis in upper Yangtze River.A total of 341 individuals were collected from 9 locations,including Banan,Jiangjing,Hejiang,Luzhou,Nanxi sections in upper Yangtze River and Qianwei section in Min River,and Qiaojia,Ningnan and Panzhihua sections in Jinsha River.The results showed that 80 polymorphic sites and 65 unique haplotypes were identified among 341 Cyt b sequences,and the average haplotype diversity(H d)and average nucleotide diversity(P i)were 0.845 and 0.007,respectively.Among 109 COI sequences,31 polymorphic sites and 23 different haplotypes were identified,and H d and P i were 0.774 and 0.005,respectively.Hierarchical analysis of molecular variance(AMOVA)showed that G.sinensis has significant genetic differentiation among sampled populations(F ST>0.5).The reconstructed neighbor-joining(NJ)phylogenetic trees,and network structure all displayed three clades of haplotypes with high resolution.The results of genetic diversity and population structure suggested that three evolutionary significant units(ESU)should be determined at least for G.sinensis population in upper Yangtze River,and it should be protected,respectively.
