摘要 目的了解贵州省新型冠状病毒肺炎(coronavirus disease 2019,COVID-19)的感染情况和所用试剂对2019新型冠状病毒核酸的检测情况。方法对五类人员(疑似病例、与确诊病例的密切接触者、每天发热门诊就诊的发热患者、湖北省武汉市等重点疫区返黔的居家隔离和集中隔离人员、每天由湖北省武汉市等重点疫区新的来黔返黔人员)进行了新型冠状病毒核酸检测,所用试剂为A和B两组,涉及样本类型有痰液、鼻拭子、咽拭子、血浆和血清。结果自2020年2月6日至2020年2月19日,完成了8500例新型冠状病毒核酸样本检测,共检测出8例阳性标本(7例为咽拭子、1例为咽拭子及痰液标本),试剂B出现未检测出ORF 1ab基因的情况,且不管是检测N或ORF 1ab基因,试剂A的CV均较小。从扩增曲线来看,试剂A的扩增曲线较试剂B典型。在痰液和咽拭子中,ORF 1ab和N基因均呈现明显的扩增曲线,但痰液中这两个靶基因的CT值均低于咽拭子。结论通过核酸检测能及早发现2019-nCoV的感染者;部分厂家2019-nCoV核酸检测试剂盒的批内重复性有待提高。 Objective To investigate the infection of Coronavirus Disease(COVID-19)in Guizhou province and the detection of 2019 novel coronavirus with the reagents used.Methods We tested five groups of people,including suspected cases,people in close contact with confirmed cases,febrile patients treated in outpatient clinics every day,home isolation and centralized isolation personnel returning to Guizhou from key epidemic areas such as Wuhan,Hubei province,and new personnel returning to Guizhou from key epidemic areas such as Wuhan,Hubei province,every day,for the 2019-nCoV.The reagents used were A and B,and the design sample types included sputum,nose swab,throat swab,serum and plasma.Results Since February 6th,2020 to February 19th,2020,we completed the detection of 8500 novel coronavirus nucleic acid samples,and a total of 8 positive(7 cases were pharyngeal swabs,and 1 case was pharyngeal swab and sputum specimen)samples were detected.The ORF 1ab gene was not detected in reagent B,and the CV of reagent A was smaller than reagent B whether it was tested for N or ORF 1ab gene.The amplification curve of reagent A was more typical than that of reagent B.In sputum and pharynx swabs,both ORF 1ab and N genes showed significant amplification curves,but the CT values of the two target genes in sputum were lower than those in pharynx swabs.Conclusion Nucleic acid testing could be used for the detection the infection of 2019-nCoV at early stage and the in-batch repeatability of 2019-nCoV nucleic acid testing kits of some manufacturers needs to be improved.
