摘要
目的:研究并建立测定烟酰胺核糖含量的离子对高效液相色谱分析法,为烟酰胺核糖质量控制提供更科学、方便的研究方法。方法:采用C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-缓冲液(pH 3.5,含0.1%庚烷磺酸钠,20:80,v/v)为流动相,流速为1 mL/min,等度洗脱,柱温为30℃,检测波长为261 nm。结果:酰胺核糖样品在1~500μg/mL的浓度范围内线性良好(相关系数为R2=0.9998,n=7),方法精密度的RSD为0.63%,回收率为99.5%(n=9)。结论:本文所建立的离子对HPLC法精密、准确,操作简便,可用于烟酰胺核糖的含量测定。
Objective:Research and establish an ion-pair high-performance liquid chromatography method for determining the content of nicotinamide ribose to provide a more scientific and convenient research method for the quality control of nicotinamide ribose.Methods:A C18 column(250 mm×4.6 mm,5μm)was used,with methanol-buffer(pH 3.5,containing 0.1%sodium heptane sulfonate,20:80,v/v)as the mobile phase,and the flow rate was 1 mL/min,isocratic elution,column temperature 30°C,detection wavelength 261 nm.Results:The amide ribose sample has a good linearity in the concentration range of 1μg/mL-500μg/mL(correlation coefficient is R2=0.9998,n=7),the method precision RSD is 0.63%,and the recovery rate is 99.5%(n=9).Conclusions:The ion pair HPLC method established in this paper is precise,accurate,and easy to operate,and can be used for the determination of nicotinamide ribose.
