摘要
目的研究信号转导子和转录激活子3(STAT3)信号通路在阿司匹林减轻缺氧诱导人滋养细胞HTR-8/SVneo损伤中的作用。方法培养人滋养细胞HTR-8/SVneo并分组。对照组进行常规处理;缺氧组进行缺氧处理;阿司匹林高、低剂量组在缺氧的基础上分别给予不同剂量的阿司匹林处理。检测细胞的增殖活力、侵袭活力及增殖基因[细胞周期蛋白D1(cyclinD1)、生存素(survivin)]、侵袭基因[基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)]和磷酸化信号转导子和转录激活子3(p-STAT3)的蛋白表达量。结果与对照组比较,缺氧组细胞的增殖活力的吸光度值(A490)、侵袭细胞数量及细胞中cyclinD1、survivin、MMP-2、MMP-9和p-STAT3的蛋白表达量均明显降低,差异有统计学意义(P<0.05);与缺氧组比较,阿司匹林高、低剂量组细胞的增殖活力A490值、侵袭细胞数量及细胞中cyclinD1、survivin、MMP-2、MMP-9和p-STAT3的蛋白表达量均明显增加,差异有统计学意义(P<0.05)。结论阿司匹林能减轻缺氧诱导人滋养细胞HTR-8/SVneo损伤,且该作用与激活STAT3信号通路有关。
Objective To investigate the role of signal transducer and activator of transcription 3(STAT3)signaling pathway in aspirin alleviating hypoxia-induced human trophoblasts HTR-8/SVneo injury.Methods Human trophoblastic HTR-8/SVneo cells were cultured and grouped.The control group was treated with routine treatment,the hypoxia group was treated with hypoxia,the aspirin low-dose group and the aspirin high-dose group were treated with different doses of aspirin on the basis of hypoxia.Proliferative activity,invasive activity and the expression of proliferative genes(cyclinD1 and survivin),invasive genes[matrix metalloproteinase 2(MMP-2),matrix metalloproteinase 9(MMP-9)]and phosphorylation signal transducer and activator of transcription 3(p-STAT3)were detected.Results Compared with the control group,the proliferation viability A490 value,the number of invasive cells and the expression of cyclinD1,survivin,MMP-2,MMP-9 and p-STAT3 in cells of hypoxic group were significantly decreased,the difference is statistically significant(P<0.05).Compared with the hypoxic group,the proliferation viability A490 value,the number of invasive cells and the expression of cyclinD1,survivin,MMP-2,MMP-9 and p-STAT3 in cells of the aspirin low-dose group and the aspirin high-dose group were significantly increased,the difference is statistically significant(P<0.05).Conclusion Aspirin can alleviate hypoxia-induced human trophoblasts HTR-8/SVneo injury and the effect is related to the activation of STAT3 signaling pathway.
