摘要
目的:探索一种可靠、有效的测定小麦籽粒单宁含量的方法,为进一步研究单宁对小麦种子生理及品质的影响提供参考。方法:应用正交试验对小麦单宁含量测定方法的进行了优化,并在56份不同基因型小麦种质资源中进行验证。结果:结果表明,经济高效的香草醛法测定小麦籽粒单宁含量的条件为:料液比为1∶10,浸提时间为6 h,离心时间为10 min,蒸馏时间为8 min,甲醇体积为2.5 mL,清洗时间为8 min,显色温度为20℃;验证试验RSD均小于5%。结论:该优化方法经济、可靠,能有效测定小麦籽粒单宁含量。
Objective:To explore a reliable and effective method for the determination of tannin content in wheat grains,and to provide a theoretical basis for the further study of the effects of tannin on wheat seed physiology and quality.Methods:The orthogonal test was used to optimize the determination method of wheat tannin content,and it was verified in 56 wheat germplasm resources of different genotypes.Results:The results showed that the optimum conditions of vanillin method were comfirmed,the ratio of material to liquid was 1∶10,the extraction time was 6 h,the centrifugation time was 10 min,the distillation time was 8 min,and the volume of methanol was 2.5 mL,the cleaning time was 8 min,the color developing temperature was 20℃.The RSD of the validation test was less than 5%.Conclusion:It is demonstrated the optimization method was economic and reliable,and it could effectively determine the tannin content of wheat grain quantity.
