没食子酸对胶原蛋白膜的改性研究 被引量:10

摘要 采用没食子酸对胶原膜进行改性,考察了没食子酸溶液浓度、反应时间和pH对胶原膜机械性能的影响。首先,以抗张强度和断裂伸长率为评价指标,采用单因素和正交试验法优化得到没食子酸改性胶原膜的最佳反应条件为:没食子酸浓度5 g/L、pH值6和反应时间4 h。然后,通过红外光谱、机械强度、耐热性、抗水性和抗氧化性测试,表征并比较了胶原膜改性前后的结构和性能变化。结果表明,没食子酸改性后胶原的三股螺旋结构未被破坏;改性胶原膜的抗张强度提高了16.1%,热变性温度提高了12.4%,吸水率降低了15.1%,并且胶原膜的超氧自由基和羟基自由基清除能力均大大提高,表现出更优的抗氧化性。 Collagen membrane was modified by gallic acid(GA), in which the effects of GA concentration, reaction time and pH were investigated on the mechanical property of collagen membrane. Firstly, single factor and orthogonal experiments were conducted in sequence by using the tensile strength and elongation at break of collagen membrane as the evaluation indexes, and the optimal GA modification conditions were obtained as the following: 5 g/L of GA concentration, pH 6.0 and 4 h of reaction time. Then, using the unmodified collagen membrane as the control, the changes in structure and property of collagen membrane modified by GA were characterized by various tests including FI-IR, tensile strength, elongation at break,thermal stability, water resistance and antioxidant activity. Results indicated that the modification by GA could not lead to the damage of triple helix structure of collagen. Compared to the control, the tensile strength of GA-modified collagen membrane was increased by 16.1% and its thermal denaturation temperature was improved by 12.4%, while its water absorption was decreased by 15.1%. In addition, GA-modified collagen membrane exhibited the greatly improved scavenging capacities for both superoxide and hydroxyl radicals as well as antioxidant activity.
