痤疮合剂对痤疮家兔耳根淋巴结朗格汉斯细胞影响实验研究 被引量:3

摘要 目的该研究旨在观察痤疮合剂对兔耳痤疮模型耳根淋巴结朗格汉斯细胞抗原提呈功能的影响。方法24只新西兰大耳白兔随机分为4组:正常组,模型组,痤疮合剂组,阳性组。正常组不做任何处理,其它3组用Kligman法制备兔耳痤疮模型,经组织学判定造模成功后,痤疮合剂组喂服痤疮合剂30 d(17.52 g·kg^-1·d^-1);阳性组喂服清热暗疮胶囊30 d(0.12 g·kg^-1·d^-1);正常组与模型组喂服等容量生理盐水。病理观察兔耳组织苏木精-伊红(HE)染色变化;流式细胞术测定耳根部淋巴节中CD207、MHC-Ⅱ、CD86、OX40阳性细胞百分率。结果痤疮合剂能有效减轻兔耳表皮层的厚度、脓肿、结节,和兔耳毛囊开口收缩及扩张的程度,抑制毛囊漏斗内角质化物质的堆积,降低真皮炎症细胞的增生和浸润;与正常组比较,其余各组耳根部淋巴结中CD207、MHC-Ⅱ、CD86、OX40阳性细胞均显著升高(P<0.05);与模型组相比,痤疮合剂组和阳性组耳根部淋巴结中CD207、MHC-Ⅱ、CD86、OX40阳性细胞百分率均降低(P<0.05)。结论痤疮合剂能通过调控痤疮模型家兔耳根淋巴结中朗格汉斯细胞抗原提呈功能,抑制Th2细胞诱导的慢性炎症,达到调节和改善兔耳痤疮模型皮损病理表现的干预效果。 Objective To study and observe the effect of Acne Mixture on the antigen-presenting function of Langerhans cells in ear root lymph nodes of rabbit acne models.Methods Twenty-four New Zealand white rabbits were randomly divided into four groups:control group,model group,Acne Mixture group,positive group.The control group did not receive any treatment,other three groups used Kligman method to prepare rabbit ear acne model.After the model was successfully established,the Acne Mixture decoction group was given Acne Mixture decoction for 30 days(17.52 g·kg^-1·d^-1);the positive group was fed with Qingre Anchuang Capsule for 30 d(0.12 g·kg^-1·d^-1).Control group and model group were fed saline of equal volume.The changes of HE staining in rabbit ear tissues were observed.The ratio of CD207,MHC-Ⅱ,CD11c and CD86 positive cells in lymphoid node of ear root was determined by Flow Cytometry.Results Acne Mixture can effectively reduce the thickness of rabbit ear epidermis,abscesses,nodules,and the extent of the rabbit ear hair follicle opening contraction and expansion,inhibit the accumulation of keratinizing substance in the hair follicle funnel,reduce the proliferation and infiltration of dermal inflammatory cells.Compared with the control group,the positive cells of CD207,MHC-Ⅱ,CD86 and OX40 in the lymph nodes of the other groups were significantly increased(P<0.05).Compared with the model group,the ratios of CD207,MHC-Ⅱ,CD86 and OX40 positive cells in the ear root lymph nodes of the Acne Mixture group and the positive group were significantly decreased(P<0.05).Conclusion Acne Mixture decoction can regulate the antigen-presenting function of Langerhans cells in the ear root lymph nodes of the rabbit model,inhibit the chronic inflammation induced by Th2 cells,and achieve the intervention effect of regulating and improving the pathological manifestations of skin lesions in the rabbit ear acne model.
