摘要
目的初步探究双氢青蒿素对肝癌细胞Huh7和Huh6生长的影响及可能存在的机制。方法培养肝癌细胞Huh7和Huh6,使用CCK-8法和结晶紫染色检测双氢青蒿素对肝癌细胞Huh7和Huh6生长抑制情况。Western Blot检测药物作用于细胞48 h后,AKT/mTOR信号通路相关蛋白的表达水平。结果与对照组相比,双氢青蒿素(12.5μM、25μM、50μM)可以下调AKT/mTOR信号通路p-AKT、p-S6K、p-S6蛋白的表达水平,从而影响肝癌细胞的生长。结论双氢青蒿素通过抑制AKT/mTOR信号通路从而抑制肝癌细胞生长。
Objective To study inhibiting effect of dihydroartemisinin(DHA)on proliferation of hepatocellular carcinoma cells and preliminary explore the mechanism.Methods Huh7 and Huh6 cells were cultured.The viability of the cells was detected with CCK-8 assay and colony forming assay.The effect of DHA on Huh7 and Huh6 migration of hepatocellular carcinoma cells was observed by using cell scratch test.We observed the changes of AKT/mTOR signaling pathway related proteins expressions with Western Blot.Results DHA(12.5μM,25μM,50μM)could significantly inhibit the proliferation of hepatocellular carcinoma cells at 48 hours.Western blot showed that compared with the control group,the DHA group could down-regulate the expressions of AKT/mTOR signaling pathway related proteins,such as p-AKT,p-S6K and p-S6,thereby affecting the growth of liver cancer cells.Conclusion DHA inhibits the growth of hepatocellular carcinoma cells by inhibiting AKT/mTOR signaling pathway.
