摘要
目的观察电针对脊髓损伤小鼠PTEN/mTOR信号通路相关蛋白表达的影响。方法64只C57BL/6小鼠随机分为假手术组、脊髓损伤组、针刺组和电针组(疏密波2/100 Hz,强度0.2 mA,15 min),每组16只,选择督脉大椎、命门穴进行治疗,1次/d,治疗28 d,免疫荧光,蛋白印迹法测定脊髓PTEN、pmTOR蛋白表达。结果与脊髓损伤组同期比较,电针组3、7、14 d小鼠脊髓中PTEN的表达降低(P<0.05,P<0.01,P<0.01)。针刺组3、7、14 d小鼠脊髓中PTEN的表达降低(P<0.05,P<0.05,P<0.05);电针组3 d、7 d、14 d、28 d小鼠脊髓中mTOR磷酸化水平增高(P<0.01,P<0.05,P<0.01,P<0.01);针刺组3 d、7 d、28 d小鼠脊髓中mTOR磷酸化水平增高(P<0.01,P<0.05,P<0.05)。结论督脉电针可促进脊髓损伤小鼠pmTOR水平,抑制PTEN蛋白的表达。
Objective To observe the effects of electro-acupuncture(EA)on the expression of proteins in PTEN/pmTOR signal pathway of spinal cord injury(SCI)mice.Methods Sixty female C57BL/6 mice were randomly divided into sham group,SCI group,acupuncture(AP)group and EA group(dense-disperse wave of 2/100 Hz frequency,0.2 mA intensity,15 minutes),15 mice in each group.Dazhui(DU14)and Mingmen(DU4)in Du meridian were selected.EA and AP were performed every day and continued for 28 days.The expressions of PTEN,phosphorylation of mTOR were determined by Immunofluorescence and Western blot.Results Compared with the SCI group,PTEN levels were decreased in the EA group at d3,d7,d14(P<0.05,P<0.01,P<0.01)and the same to that of the AP group(P<0.05,P<0.05,P<0.05).The phosphorylation of mTOR was increased in the EA group at d3,d7,d14,d28(P<0.01,P<0.05;P<0.01,P<0.01)and increased in the AP group at d3,d7,d28(P<0.01,P<0.05;P<0.05).Conclusion EA at Du meridian may promote the phosphorylation of mTOR and inhibit the expression of PTEN.
