不同浓度黄芪提取物对高糖损伤HUVECs的保护机制

摘要 目的:研究不同浓度的黄芪提取物对HUVECs损伤的保护机制。方法:将HUVECs细胞分为正常组、高糖组、高渗组等12组分别培养12h、24h、48h,考察HUVECs细胞活力;观察不同浓度黄芪提取物干预后,HUVECs细胞中ET-1、eNOS、p-eNOS的活力以及mRNA和蛋白量的表达情况。结果:MTT检测结果显示高糖组会显著影响HUVECs细胞的活力,随着培养时间的延长,黄芪提取物各组对高糖损伤的保护不断增强(P<0.05)。PCR、Western blot法检测结果显示,与正常组相比,高糖组的ET-1表达明显升高(P<0.05),eNOS表达量显著降低(P<0.05);3种不同浓度的黄芪提取物均能抑制ET-1的表达,增加eNOS表达;Western blot法检测eNOS结果显示高糖、黄芪提取物对eNOS蛋白表达无影响(P>0.05)。结论:高糖组能显著抑制HUVECs细胞的活性,黄芪提取物组可不同程度地改善高糖抑制作用(P<0.05),且黄芪提取物高浓度组保护作用最强,说明不同浓度的黄芪总黄酮提取物能从调节ET-1、eNOS、p-eNOS表达量方面对高糖损伤的HUVECs产生保护作用。 Objective:To Study on the protective mechanism of different concentration extracts from astragalus flavonoids in high glucose damaged HUVECs human umbilical vein endothelial cells(HUVECs).Methods:The HUVECs cells were divided into 12 different groups and given with glucose,mannitol and astragalus extracts in three concentrations,evaluated the protection by measuring the activity,mRNA and Protein expressionabout about ET-1,eNOS,p-eNOS.Results:MTT test show cell activity of the high glucose group was obviously lower than that of normal control group,as the time went on the cell activities of astragalus extractswith different concentration groups were significant;PCR,Western blot test show comparing with normal group,the secretion and the expression of ET-1 with a significant rise(P<0.05),the secretion and the expression of eNOS with a significant decrease(P<0.05);Three different concentrations of astragalus extracts could significantly inhibit the expression of ET-1 and increase the expression of eNOS.Western blot test also showed the expression of total eNOS had no significant change(P>0.05).Conclusion:The high glucose groups can significantly inhibit the activity of HUVECs cells,and the astragalus extract group can improve the inhibition of high glucose to varying degrees(P<0.05);Astragalus extract high concentration groups of Astragalus extract have the strongest protective effect;the results show different concentrations of Astragalus extracts can protect HUVECs with high glucose damage in et-1,eNOS and p-enos ways.
