摘要
采用纤维素酶法提取黑豆中的可溶性膳食纤维。以可溶性膳食纤维提取率为指标,通过单因素试验,Plackett-Burman因素筛选,结合正交试验设计对提取工艺进行优化。评价了料水比、酶添加量、酶解温度、酶解pH和酶解时间5个因素对可溶性膳食纤维提取效果的影响。确定最优提取工艺为料水比1∶25(g/mL)、酶添加量2.2%、酶解温度45℃、酶解时间7 h、酶解pH 5.0,在此条件下,黑豆中可溶性膳食纤维的提取率为38.40%。
Soluble dietary fiber was extracted from black soybean by cellulase method.With the extraction rate of soluble dietary fiber as index,through the single factor experiments,the Plackett-Burman factor screening and the orthogonal experimental design,the extraction process was optimized.The influence of 5 factors(ratio of material to water,addition of cellulase,enzynolysis temperature,enzynolysis time and enzynolysis pH)on extraction rate of soluble dietary fiber were evaluated.The optimal extraction conditions were as follows:the ratio of material to water 1∶25(g/mL),the addition of cellulase 2.2%,the enzynolysis temperature 45℃,the enzynolysis time 7 h,the enzynolysis pH 5.0.Under the conditions,the extraction rate of soluble dietary fiber in black bean was 38.40%.
