广霍香组培工厂化育苗技术体系研究 被引量:5

摘要 为打破广藿香传统扦插繁殖,为生产上提供大量优质种苗,以收集的广藿香优株嫩叶为外植体,研究影响芽诱导、增殖、壮苗生根、移栽成活率的主要因素。结果表明,培养基为MS+蔗糖3%+6-BA 0.5 mg/L+NAA 0.01 mg/L有利于诱导芽的形成、培养基为MS+蔗糖3%+6-BA 0.2 mg/L+NAA 0.01 mg/L有利于芽的增殖、壮芽伸长;1/2 MS+蔗糖1.5%+NAA 0.2 mg/L+活性炭1g/L有利于诱导芽生根;适宜基质配比泥炭土∶蛭石∶河沙为7∶2∶1(体积比),于晚春和秋季移栽,移栽的成活率最高,达93%左右。以此为基础研究出一套组织培养工厂化育苗的生产技术路线。本研究为广藿香的大量生产提供了可行的依据基础。 In order to change the traditional cuttage propagation and provide a large number of high-quality planting materials for production of Pogostemon cablin fresh leaves collected from P.cablin(Blanco)Benth were used as explants to regenerate plants,and the key factors influencing shoots induction,proliferation,rooting and transferring survival rate were analyzed.The results indicated that the suitable mediums for culture of the leaf explants were MS+3%sucrose+BA 0.5 mg·L^-1+NAA0.01 mg·L^-1 for shoot induction,MS+3%sucrose+BA 0.2 mg·L^-1+NAA0.01 mg·L^-1 for shoot proliferation and elongation,and 1/2 MS+1.5%sucrose+NAA 0.2 mg·L^-1+1 g·L^-1 activated charcoal for rooting.Rooted plantlets were transferred onto the substrates in different seasons.The suitable substrate was a mixture of peat soil:vermiculite:sand=7:2:1,on which the rooted plantlets had the highest survival rate,about 93%when transferred in late spring or autumn.A technical route for mass propagation of P.cablin via tissue culture can hence be established based on these results.
