海南叶用甘薯茎尖脱毒再生技术体系研究 被引量:2

摘要 以海南叶用甘薯顶芽茎尖生长点为外植体,对海南叶用甘薯脱毒苗技术进行研究,探讨不同浓度6-BA对叶用甘薯茎尖启动培养的影响及基础培养基浓度、糖浓度及激素浓度对叶用甘薯组培苗生长的影响。结果表明:海南叶用甘薯茎尖启动培养最适培养基为MS+蔗糖25 g/L+卡拉胶8 g/L+6-BA 1.0 mg/L;基础培养基为MS,蔗糖浓度为55 mg/L,IBA为0.2 mg/L,不添加KT较适合继代繁殖。获得脱毒苗后在MS+糖55 g/L+IBA 0.2 mg/L+卡拉胶8 g/L上继代培养,30 d后长到15 cm左右进行炼苗移栽。炼苗移栽至用1000倍多菌灵消毒后的基质上(椰糠∶菜园土∶珍珠岩=1∶3∶1,体积比),10 d成活率90%,植株恢复生长,开始萌发新叶,30 d成活率为85%左右。通过反复几次实验,该方案较稳定适合海南叶用甘薯组培脱毒苗的生产和应用。 To establish the rapid propagation technology system by using the terminal bud explants of sweet potato stem tips.This article explores the effects of different 6-BA concentrations on the shoot tip start up culture of sweet potato.The effects of different basic medium concentrations,sugar concentrations and hormone concentrations on the growth of sweet potato tissue culture plant let were tested.The results showed that the most suitable basal medium was MS,and the most suitable shoot tip induction formula was MS+sucrose 25 g·L^-1+carrageenan 8 g·L^-1+6-BA1.0mg·L^-1.The addition of 55mg·L^-1 sugar and 0.2mg·L^-1IBA could effectively promote the growth of Ipomoea Batatas(L.)Lam plant lets.The addition of KT did not promote the growth of sweet potato tissue culture plant lets.These virus-free leaves were inoculated with I.Batatas plant lets and grown on MS+sugar 55 g·L^-1+IBA 0.2 mg·L^-1+carrageenan 8 g·L^-1.After 30 days of subculture,they can grow to about 15 cm,and then will be going to transplant.The cultivation substrate was sterilized with 1000 fold carbendazim.After 10 days of transplantation,the survival rate was 90%,and the plants began to resume growth and germinate new leaves;at 30 days,the survival rate was about 85%.After several experiments,this scheme is more stable and suitable for the production and application of I.Batatas virus-free plant lets in Hainan.
