脂联素对乳腺癌小鼠血管内皮生长因子和基质金属蛋白酶-9表达的影响

Effect of adiponectin on the VEGF and MMP-9 expression in breast cancer mice

目的探讨脂联素对乳腺癌小鼠血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)表达的影响。方法选取80只健康雌性小鼠,采用含有二甲基苯蒽的芝麻油建立乳腺癌模型,成瘤率50%,建模成功后依据随机数字表将小鼠分为2组,模型组20只给予常规饲养,脂联素组20只给予重组鼠脂联素蛋白注射液腹腔注射,连续7 d。分别采用酶联免疫吸附法和Western blot(WB)法检测2组小鼠造模成功后和造模成功1,2,3周后血清VEGF、MMP-9水平和肿瘤组织中VEGF、MMP-9表达水平,造模成功3周后处死小鼠称肿瘤质量并计算瘤体积。结果脂联素组造模成功1,2,3周后血清VEGF、MMP-9水平及肿瘤组织中VEGF、MMP-9表达水平均明显低于同组造模成功后和同期模型组(P均<0.05);脂联素组肿瘤质量明显低于模型组(P<0.05),肿瘤体积明显小于模型组(P<0.05)。结论脂联素可能通过抑制乳腺癌小鼠VEGF、MMP-9表达而抑制病情进展。

Objective It is to discuss the effect of adiponectin on the vascular endothelial growth factor(VEGF)and matrix metalloproteinase-9(MMP-9)expression in breast cancer mice.Methods Eighty healthy mice were selected to establish breast cancer models with sesame oil containing dimethylbenzanthracene.After successful modeling,they were divided into 2 groups according to the random number table,the model group(n=20)was normally fed,while the adiponectin group(n=20)was treated with recombinant adiponectin protein injection by intraperitoneal injection for 7 days.The levels of serum VEGF and MMP-9 and the levels of VEGF and MMP-9 expressions in tumor tissue were respectively detected by enzyme-linked immunosorbent assay and Western blot(WB)after modeling and at 1,2,3 weeks after modeling.3 weeks after successful modeling,the mice were sacrificed to weigh the tumor mass and calculate the tumor volume.Results The levels of serum VEGF and MMP-9 and the levels of VEGF and MMP-9 expressions in tumor tissue after 1,2 and 3 weeks of culture in the adiponectin group were significantly lower than those before the culture and in the model group at the same period(P<0.05).The volume and weight of tumors in adiponectin group were significantly lower than those in model group(P<0.05).Conclusions Adiponectin may inhibit the progression of breast cancer by inhibit the VEGF and MMP-9 expression in breast cancer mice.
