金针菇类扩张蛋白fvexpl2鉴定及其表达模式分析 被引量:1

Identification and Experssion Patterns Analysis of Expansin fvexpl2 in Flammulina filiformis

摘要 基于金针菇基因组与转录组数据,利用在线软件BLAST鉴定了一个扩张蛋白类似物编码基因,命名为fvexpl2,并对其基因结构进行了鉴定,通过构建进化树的方法分析了fvexpl2与担子菌、子囊菌扩张蛋白类似物以及植物扩张蛋白之间的亲缘关系。结果表明:扩张蛋白类似物外显子的数目为4个,内含子的数目为3个,并且CDS的长度是438 bp,分子量为15 746.88 Da,等电点为4.93;进化树图显示fvexpl2与担子菌类扩张蛋白处于同一亚支,亲缘关系较近。实时荧光定量PCR结果则表明fvexpl2在菌柄伸长部位具有较高的表达水平,其表达水平远远高于菌盖、原基以及菌丝部位的表达水平,且其在伸长较快部位表达水平显著高于伸长较慢部位表达水平,所以fvexpl2可能对菌柄伸长起重要作用。 Based on the genome and transcriptome data of Flammulina filiformis,this experiment identified an expansin analog encoding gene using online software BLAST.It was named fvexpl2 and identified its gene structure.The phylogenetic relationship between fvexpl2 and basidiomycetes,ascomycetes and plants was analyzed by constructing evolutionary tree.The results showed that four exons and three introns contained in the dilatant protein analogue,and the length of coding sequence was 438 bp,molecular weight 15 746.88 Da,isoelectric point 4.93.The construction of evolutionary tree showed that fvexpl2 and basidiomycetes were in the same subclan and had close relationship.According to the real-time fluorescence quantitative PCR results,we could understand that fvexpl2 was much higher than that in the cap,primordium and mycelium,and it had a higher expression level in the fast elongation region of the stipe compared with the slow elongation region.Therefore,fvexpl2 might play an important role in the elongation of the stipe.
