改良苏木精-伊红染色在病理诊断中的应用研究 被引量:1

Application of modified hematoxylin-eosin staining in pathological diagnosis

摘要 目的研究改良苏木精-伊红(hematoxylin-eosin,HE)染色在病理诊断中的应用价值。方法选择丽水市妇幼保健院2018年1月至2019年1月行病理检查的石蜡切片组织标本500份,分别对石蜡切片组织标本进行常规HE染色(对照组)、改良HE染色(观察组),比较两组染色优片率、染色质量评分,并比较两种染色方法病理检查对各疾病的诊断结果。结果观察组胃肠道活检组织、骨组织、腹腔脏器组织、甲状腺活检组织的染色优片率分别为95.51%(85/89)、94.87%(111/117)、96.74%(208/215)、94.94%(75/79),均高于对照组的82.02%(73/89)、85.47%(100/117)、91.16%(196/215)、83.54%(66/79)(χ^2=8.114、5.834、5.895、5.339,均P<0.05)。观察组各类标本的组织结构清晰度、细胞核染色效果、细胞质染色效果、细胞核质对比效果评分均高于对照组(t=7.146、7.301、7.499、7.566,8.062、8.225、8.287、8.250,10.954、11.044、10.786、11.305,10.941、11.222、10.996、10.750,均P<0.05)。观察组对胃肠道病变、骨病、腹腔脏器病变、甲状腺病变的诊断准确率分别为96.63%(86/89)、96.58%(113/117)、98.14%(211/215)、96.20%(76/79),均高于对照组的84.27%(75/89)、88.89%(104/117)、93.49%(201/215)、86.08%(68/79)(χ^2=7.869、5.138、5.798、5.016,均P<0.05)。结论改良HE染色在病理诊断中具有良好的应用价值,可提高石蜡切片组织标本的染色质量,有利于提高病理诊断的准确性。 Objective To study the value of hematoxylin-eosin(HE)staining in pathological diagnosis.Methods From January 2018 to January 2019,500 paraffin section specimens were selected from the Maternal and Child Health Hospital of Lishuifor pathological examination.The routine HE staining(control group)and modified HE staining(observation group)were performed on paraffin section specimens respectively.The excellent staining rate and staining quality score of the two groups were compared,and the diagnostic results of pathological examination of the two staining methods for each disease were compared.Results In the observation group,the optimal staining rates of gastrointestinal biopsy tissue,bone tissue,abdominal viscera tissue and thyroid biopsy tissue were 95.51%(85/89),94.87%(111/117),96.74%(208/215)and 94.94%(75/79),respectively,which were higher than those in the control group[82.02%(73/89),85.47%(100/117),91.16%(196/215),83.54%(66/79)](χ^2=8.114,5.834,5.895,5.339,all P<0.05).The clarity of tissue structure,nuclear staining effect,cytoplasmic staining effect and nuclear/cytoplasmic contrast effect score of the observation group were all higher than those of the control group(t=7.146,7.301,7.499,7.566,8.062,8.225,8.287,8.250,10.954,11.044,10.786,11.305,10.941,11.222,10.996,10.750,all P<0.05).The diagnostic accuracy of gastrointestinal lesions,bone diseases,abdominal viscera lesions and thyroid lesions in the observation group were 96.63%(86/89),96.58%(113/117),98.14%(211/215)and 96.20%(76/79),respectively,which were higher than those in the control group[84.27%(75/89),88.89%(104/117),93.49%(201/215),86.08%(68/79)](χ^2=7.869,5.138,5.798,5.016,all P<0.05).Conclusion Modified HE staining has good application value in pathological diagnosis.It can improve the staining quality of paraffin sections and the accuracy of pathological diagnosis.
