摘要
目的观察柚皮苷联合低强度脉冲式超声波对兔膝关节软骨缺损的修复作用。方法24只新西兰大耳白兔建立双侧膝关节软骨缺损模型后随机均分为四组:A组予安慰剂灌胃联合假脉冲处理;B组予柚皮苷汤10 mL/d灌胃联合假脉冲处理;C组予安慰剂灌胃联合低强度脉冲式超声波刺激膝关节缺损处,每日1次,每次20 min;D组予柚皮苷汤10 mL/d灌胃联合低强度脉冲式超声波刺激。8周后处死各组动物,肉眼观察膝关节缺损软骨处变化,软骨组织病理切片采用甲苯胺蓝染色观察糖胺聚糖分泌情况,采用国际软骨修复学会(ICRS)评分评估软骨修复效果。结果 8周后,A组软骨缺损处未见肉芽组织,缺损面积较大;B、C、D组可见不同程度的肉芽组织生长,且缺损面积较小,其中D组缺损部位组织生长与周围正常组织最接近。A、B、C组甲苯胺蓝染色后细胞较少,间充质染色浅;D组细胞较大,间充质染色均匀,软骨深层染色深。D组ICRS评分高于A、B、C组(P<0.01)。结论柚皮苷联合低强度脉冲式超声波能提高兔膝关节软骨缺损的修复质量。
Objective To observe the effect of naringin combined with low-intensity pulsed ultrasound(LIPU) on the repair of knee joint cartilage defects in rabbits.Methods After establishment of bilateral knee joint cartilage defect model,24 New Zealand big-eared white rabbits were randomly divided into 4 groups with 6 rabbits each,which were treated with placebo gavage plus false pulse treatment(group A),naringin decoction 10 mL/d gavage plus false pulse treatment(group B),placebo gavage plus LIPU treatment(stimulation of knee joint defects 20 minutes per day)(group C),and naringin decoction 10 mL/d gavage plus LIPU treatment(group D),respectively.After 8 weeks of intervention,the animals in each group were killed,and the changes of knee joint cartilage defects were observed.Glycosaminoglycan secretion was observed by toluidine blue-stained pathological section of cartilage tissues.International Cartilage Repair Society(ICRS) score was used to evaluate the effect of cartilage repair.Results After 8 weeks,no granulation tissue was found in the cartilage defect and the defect area was larger in group A.Different degrees of granulation tissue growth were found and the defect area was smaller in groups of B,C and D.It was difficult to distinguish the defect repair part from the surrounding normal cartilage in group D.There were fewer cells and the toluidine blue staining was light in groups of A,B and C.The cells were larger,the toluidine blue staining was uniform,and the staining of deep cartilage tissue was heavy in group D.The ICRS score in group D was higher than that in groups of A,B and C(P<0.01).Conclusion Naringin combined with LIPU can improve the quality of repair of knee joint cartilage defects in rabbits.
