牡丹废弃物用于制备发酵可利用还原糖的研究 被引量:4

摘要 本研究以牡丹废弃物(叶、果荚、枝)为研究对象,采用傅里叶变换红外(FTIR)光谱分析其木质纤维素种类,分别采用微波-碱法和水浴-碱法预处理原材料后,用GC-MS结合FTIR光谱分析不同预处理方法对原材料的处理效果,并采用纤维素酶解法制备还原糖进行还原糖产量分析,以评价牡丹废弃物作为原材料制备发酵可利用还原糖进行生物转化的潜能。结果表明,牡丹不同部位废弃物的木质纤维素含量不同,牡丹枝中含量最高,综纤维素含量是枝>果荚>叶;木质素为SG型(即愈创木基和紫丁香基木质素),以愈创木基木质素为主。微波-碱法和水浴-碱法预处理均能有效去除愈创木基,破坏部分半纤维素,提高纤维素酶解产糖效率,差异不明显;预处理也能有效去除原材料中的可溶性成分,提高还原糖产量;以牡丹果荚为原料的还原糖产量比以枝和叶为原料的产糖量至少提高46.62%,因此,牡丹果荚更适合作为制备发酵可利用还原糖的原材料。 In this study,the lignocellulose composition of Paeonia ostii wastes( containing leaves,fruit pods,branches) was analyzed by the Fourier transform infrared( FTIR) spectrometer.After being treated with alkaline under microwave irradiation or water bath,the pretreated biomass was evaluated using GC-MS combined with FTIR analysis.Meanwhile,the fermentable reducing sugar content was determined by the cellulase hydrolysis method to evaluate the potential of P.ostii wastes as raw material to be bio-transformed into other cost-effective products.The results showed that the lignocellulose content in different parts of P.ostii was different.Branches had the highest lignocellulose,and the content of cellulose and hemicellulose gradually decreased in branches,pods and leaves.The type of the lignin was SG( contained syringyl-lignin and guaiac-lignin) and the guaiac-lignin was the main lignin.Being treated with alkaline under microwave irradiation or water bath all could effectively remove guaiac-lignin,destroy the part of the hemicellulose,improve the enzymolysis of celluloses and the difference was not obvious.Besides,pretreatment could effectively remove the soluble components to increase the yield of reducing sugar.By comparative analysis,P.ostii pods were proved to be more suitable to produce fermentable reducing sugar with at least 46.62% higher compared with those of branches and leaves.
