摘要
目的建立盐巴戟天含量和特征图谱测定方法,考察不同蒸制时间对盐巴戟天含量及特征图谱的影响。方法采用超高效液相色谱(UPLC)法同时测定水晶兰苷含量和环烯醚萜类特征图谱、高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法测定耐斯糖含量和超高效液相色谱-电喷雾检测器(UPLC-CAD)法测定寡聚糖类特征图谱,并结合方差分析对不同蒸制时间的特征图谱进行差异性研究。结果分别建立了盐巴戟天环烯醚萜类和寡聚糖类UPLC特征图谱,确定共有特征峰分别为12个和20个;不同蒸制时间的水晶兰苷含量、耐斯糖含量及特征图谱均发生明显变化,优选盐巴戟天蒸制时间为1 h。结论实验建立的方法稳定、准确、灵敏度好,可用于盐巴戟天的质量控制。
Objective To establish a method for determination of content and characteristic chromatogram of Salt-steamed Morindae Officinalis Radix(SMOR)and to investigate the influence of different Steaming time on the content and characteristic chromatogram of SMOR.Methods UPLC method is used to simultaneously determine the contents of monotropein and characteristic chromatogram of iridoids,HPLC-ELSD method to determine the content of nystose,UPLC-CAD method to determine the characteristic chromatogram of oligosaccharides,and variance analysis to study the difference of characteristic chromatogram of different steaming time.Results The UPLC characteristic chromatograms of iridoids and oligosaccharides are established,and 12 and 20 common characteristic peaks are confirmed,respectively.There are significant change in content of monotropein and nystose and the characteristic chromatogram at different steaming times.The optimal steaming time of SMOR is 1 h.Conclusion The established method is stable,accurate and sensitive,and can be used for quality control of SMOR.
