宫颈癌细胞MFGE8表达与生物学行为的相关性

Correlation between Expression of MFGE8 and Biological Behavior in Cervical Cancer Cells

目的探讨宫颈癌细胞乳脂肪球表皮生长因子8蛋白(MFGE8)表达与生物学行为的相关性。方法SiHa细胞分为三组:对照组、实验1组与实验2组,对照组不进行转染,实验1组与实验2组分别转染siRNA1-MFGE8与siRNA2-MFGE8,采用实时荧光定量PCR检测MFGE8 mRNA表达,Western blot检测MFGE8蛋白表达,CCK-8法检测细胞增殖能力,流式细胞术检测细胞周期与细胞凋亡。结果转染24 h与48 h后,实验1组与实验2组的MFGE8 mRNA与蛋白相对表达水平、细胞增殖指数都低于对照组(P<0.05),细胞凋亡指数显著高于对照组(P<0.05),实验1组与实验2组之间对比差异无统计学意义(P>0.05)。转染后48 h,三组的细胞G1、S、G2/M期对比差异都无统计学意义(P>0.05)。结论宫颈癌细胞中MFGE8表达呈现高表达情况,抑制MFGE8能抑制宫颈癌细胞的增殖,促进其凋亡,但是对细胞周期无显著影响。

Objective To investigate the correlation between the expression of breast fat globule EGF factor 8(MFGE8 and biological behavior in cervical cancer cells.Methods Si Ha cells were divided into 3 groups:the control group,the experiment group 1 and the experiment group 2,and the control group were not transfected,the experiment group 1 and the experiment group 2 were transfected with siRNA1-MFGE8 and siRNA2-MFGE8,respectively,the MFGE8 mRNA expression were detected by the real-time PCR,MFGE8 protein expression were detected by Western blot,cell proliferation were detected by CCK-8 method,and cell cycle and apoptosis were detected by flow cytometry.Results After transfection for 24 h and 48 h,the relative expression levels of MFGE8 mRNA and protein,the cell proliferation index in the experimental group 1 and the experimental group 2 were lower than those of the control group(P<0.05),and the apoptotic index were significantly higher than that of the control group(P<0.05).There were no significant difference compared between the experimental group 1 and the experimental group 2(P>0.05).At 48 h after transfection,there were no significant difference in G1,S,G2/M phase compared among the 3 groups(P>0.05).Conclusion The expression of MFGE8 in cervical cancer cells is highly expressed.Inhibition of MFGE8 can inhibit the proliferation of cervical cancer cells and promote their apoptosis,but has no significant effect on cell cycle.
