摘要
目的探索高原环境中母体绵羊妊娠期缺氧对胎羊心脏发育的蛋白组学影响.方法将15只妊娠期绵羊随机分为平原对照组(LAC,6只)、轻度低氧暴露组(MHE,5只)和重度低氧暴露组(SHE,5只).应用高原人工实验舱建立不同程度的妊娠期绵羊缺氧模型,建模120 d后应用iTRAQ技术检测评估胎羊心脏不同蛋白的表达情况.结果本实验共鉴定了16676个肽段和2506个蛋白质,共鉴定出80个差异表达的蛋白,其中41个蛋白表达上调,39个蛋白表达下调.不同分组间检验阈值为缺氧诱导蛋白表达水平的1.3倍以上,差异有统计学意义(P<0.05).结论本实验证实母体妊娠期缺氧能够显著影响胚胎心脏发育过程中与缺氧相关蛋白的表达,验证母体妊娠期低氧环境在子代先天性心脏损伤发生中的作用及其可能机制,为临床先天性心脏疾病的一级预防提供了借鉴.
Objective To explore the change of the fetal sheep cardiac proteomics under the exposure of hypoxia environment.MethodsSetting up the different degrees of hypoxia pregnancy sheep mode by artificial cabin and sixteen artificial insemination breading sheeps were randomly divided into control group(n=6),mild hypoxia exposure group(n=5)and severe hypoxia exposure group(n=5).iTRAQ-based quantitative proteomics was employed to identify differentially expressed proteins after modeling 120 days.Results In tissue analysis,a total of l6676 peptides and 2506 proteins were quantified.80 differentially expressed proteins were identified in fetal cardiac tissues and with 4l upregulated and 39 downregulated proteins(P<0.05).Conclusion This primary experimental results show that hypoxia can cause scrious damage to the heart development of the fetal sheep and lead to the significant changes of hypoxia-associated proteins expression,which may be used to the further investigation for clinical applications.
