摘要
本研究以小美旱杨的叶片作为外植体,对小美旱杨直接诱导不定芽进行了研究。结果表明:使用2%NaClO最佳消毒时间为8 min;小美旱杨叶片诱导不定芽最佳培养基为MS+TDZ 0.3 mg/L+IBA 0.3 mg/L;小美旱杨不定芽增殖的最佳培养基为MS+6-BA 0.5 mg/L+IBA 0.3 mg/L;小美旱杨生根的最佳培养基为1/2MS+IBA 0.3 mg/L+NAA 0.08 mg/L。本研究成功建立小美旱杨的再生体系,对小美旱杨的品种改良具有重要意义。
In this study, the leaves of Populus popularis were used as explants to directly induce adventitious buds. The results showed that the optimum sterilization time of 2% NaClO was 8 min, the optimum medium of adventitious buds induced by the leaves of P. popularis was MS +TDZ 0.3 mg/L +IBA 0.3 mg/L, and the best medium for the proliferation of adventitious buds in P. popularis was MS+6-BA 0.5 mg/L+IBA 0.3 mg/L;The best medium for the rooting of P. popularis is 1/2 MS+IBA 0.3 mg/L+NAA 0.08 mg/L. The successful establishment of the regeneration system of P. popularis in this study is of great significance to the improvement of the variety of P. popularis.
