Genetic Diversity Analysis of Mungbean Germplasm Resources From Anhui Province with Molecular Markers

摘要 为了解安徽地区绿豆种质资源的遗传背景和亲缘关系,本研究挑选了27对(15对InDel和12对SSR标记)条带清晰、多态性好的分子标记对安徽地区的66份绿豆地方品种进行遗传多样性分析。研究结果表明,单个标记检测到的等位基因数(Na)在2~4个,平均为2.57个;有效等位基因数(Ne)为1.38~3.45个,平均为2.03个;Shannon信息指数(I)的变幅介于0.45~1.31之间,平均值为0.76;Nei’s基因多样性指数在0.28~0.71之间,平均为0.49;标记的多态性信息含量(PIC)值在0.24~0.66之间,平均PIC含量为0.40。66份资源间的遗传相似系数在0.33~1.00之间,平均为0.61,UPGMA聚类分析将66份材料在遗传相似系数为0.525处分为两大类群。但聚类结果没有严格按照地理来源进行划分,来源于不同地理区域的资源被划分在一起。上述研究结果表明安徽省绿豆地方品种遗传基础狭窄,遗传多样性水平较低。因此,为丰富安徽省绿豆资源遗传多样性水平,需加强资源引进和种质创新工作。本研究结果为分析安徽地区绿豆品种亲缘关系提供理论依据,同时也为资源收集和提高优良种质利用率提供依据。 In order to explore the genetic background and genetic relationship of mungbean germplasm resources from Anhui Province, 27 molecular markers(15 InDel and 12 SSR markers) with clear band and good polymorphism were selected to analyze the genetic diversity of 66 mungbean landrace from Anhui Province. The number of alleles(Na) per marker ranged from 2~4, with an average of 2.57. The effective number of alleles(Ne) varied from1.38~3.45, with an average of 2.03. The Shannon information index(I) was between 0.45~1.31, with an average of0.76. The Nei’s gene diversity index was between 0.28~0.71, with an average of 0.49. The value of the polymorphism information content(PIC) was between 0.24~0.66, with a mean of 0.40. The genetic similarity coefficient among 66 mungbean germplasm resources ranged from 0.33~1.00, with a mean value of 0.61. The UPGMA(Unweight pair group method with arithmetic mean) clustering analysis at the genetic similarity value of 0.525 grouped the 66 materials into two clusters. However, the clustering pattern did not strictly follow their geographical origins, accessions from diverse geographical locations were grouped into same cluster. These results showed that the genetic base of mungbean landrace from Anhui Province was narrow and the genetic diversity was low.Therefore, in order to enrich genetic diversity of mungbean germplasm resources in Anhui Province, introduction and germplasm innovation should be strengthened. This study would provid a theoretical basis for eluciating the relationship of mungbean accessions from Anhui Province and lay the foundation for the collection and utilization of excellent mungbean materials.
