泰利霉素体外抗金黄色葡萄球菌生物膜活性研究 被引量:1

Study on the Activity of Telithromycin against Staphylococcus Aureus Biofilm in Vitro

摘要 目的:比较泰利霉素与常见抗菌药物(阿奇霉素、克林霉素、万古霉素和达托霉素)对金黄色葡萄球菌生物膜的作用。方法:本研究选择了2015年从华中科技大学协和深圳医院收集的4株甲氧西林敏感金黄色葡萄球菌(MSSA)和4株甲氧西林耐药金黄色葡萄球菌(MRSA),通过药物作用,使用96孔微板法定量结晶紫染色检测菌株产生物膜的能力,通过cfu计数方法对生物膜中的细菌进行定量分析。结果:在亚抑制浓度下(4×MIC),泰利霉素明显优于阿奇霉素或克林霉素抑制MSSA菌株生物被膜的形成,优于万古霉素或达托霉素抑制MRSA生物膜的形成。在8×MIC浓度下,泰利霉素优于阿奇霉素或克林霉素清除已经形成的MSSA菌株生物被膜,优于万古霉素或达托霉素清除已经形成的MRSA菌株生物被膜,同时,对已经形成的金黄色葡萄球菌生物膜中活菌的杀灭作用,泰利霉素与达托霉素作用相似,明显优于阿奇霉素、克林霉素和万古霉素。结论:泰利霉素对金黄色葡萄球菌生物膜的疗效优于阿奇霉素、克林霉素、万古霉素或达托霉素。 Objective This study aimed to explore telithromycin against Staphylococcus aureus biofilms and compared with azithromycin,clindamycin,vancomycin and daptomycin.Methods Four biofilm positive methicillin-susceptible and four biofilm positive methicillin-resistant S.aureus isolates(MSSA and MRSA,respectively)that collected from Huazhong university of science and technology union Shenzhen hospital in 2015 were used for this study.Biofilm biomasses were detected by crystal violet staining and the adherent cells in the established biofilms were quantified by CFU determination.Results The subinhibitory concentration of telithromycin(1/4×MIC)was significantly superior to azithromycin or clindamycin inhibited the biofilm formation of 3 MSSA isolates,and inhibited the biofilm formation of all MRSA isolates more effectively than vancomycin or daptomycin.Telithromycin(8×MIC)eradicated more established biofilms than azithromycin or clindamycin in the 2 MSSA isolates,and eliminated the established biofilms of 3 MRSA isolates more effectively than vancomycin or daptomycin.Telithromycin(8×MIC)killed more adherent cells in the established biofilms than azithromycin or clindamycin in all the MSSA isolates,and killed more adherent cells than vancomycin in all the MRSA isolates.Daptomycin also showed an excellent effect on the adherent cells of all the 4 MRSA isolates and was similar to that treated with telithromycin.Conclusion Telithromycin is more effectively than azithromycin,clindamycin,vancomycin or daptomycin against the biofilms of S.aureus.
