摘要
目的:探讨CtBP2对胶质瘤细胞增殖、侵袭和EMT的影响。方法:将体外培养的恶性胶质瘤细胞系(U87MG、LN229)和人正常星形胶质细胞(Astrocyte)分为转染组、对照序列组和空白组,分别转染后6小时,更换培养基。48小时后收集细胞进行Western blot、CCK-8及Transwell侵袭测定。结果:CtBP2在胶质瘤细胞中高表达;在胶质瘤中高表达的CtBP2促进胶质瘤细胞增殖,且促进其侵袭、转移;高表达的CtBP2导致了EMT的诱导,抑制了E-cadherin的表达,促进了Vimentin的表达。结论:在胶质瘤细胞中高表达的CtBP2不仅促进胶质瘤细胞增殖、侵袭并且导致了EMT的诱导。
Objective:To investigate the effect of CtBP2 on proliferation,invasion and EMT of glioma cells.Methods:U87MG,LN229 and astrocyte cells were divided into three groups:Transfection group,control sequence group,and blank group.The medium was changed 6 hours after transfection.Cells were collected 48 hours later for Western blot,CCK-8 and Transwell invasion assay.Results:CtBP2 was highly expressed in glioma cells.Highly expressed CtBP2 in glioma promoted glioma cell proliferation,invasion and metastasis.The high expression of CtBP2 led to the induction of EMT,inhibited the expression of E-cadherin,and promoted the expression of Vimentin protein.Conclusion:CtBP2,which is highly expressed in glioma cells,not only promotes the proliferation and invasion of glioma cells,but also leads to the induction of EMT.
