高肺血流性肺动脉高压大鼠血清和肺组织补体成分1q/肿瘤坏死因子相关蛋白9水平及其与血流动力学和右心室肥厚的相关性分析

摘要 目的观察高肺血流性肺动脉高压大鼠血清和肺组织补体成分1q/肿瘤坏死因子相关蛋白9(C1q/tumor necrosis factor-related protein 9,CTRP9)水平,及其与血流动力学和右心室肥厚的相关性。方法将50只成年雄性SD大鼠按照完全随机数字表法分为假手术组和分流组,每组各25只,分流组大鼠制作高肺血流性肺动脉高压模型。比较两组大鼠血流动力学指标[肺动脉平均压(mean pulmonary artery pressure,mPAP)、右心室平均压(mean right ventricular pressure,mRVP)]、右心室肥厚指数(right ventricular hypertrophy index,RVHI)、CTRP9和一氧化氮(nitric oxide,NO)水平。结果分流组大鼠mPAP、mRVP、RVHI、血清NO水平、血清和肺组织CTRP9水平均显著高于假手术组(均P<0.05)。Pearson相关性分析结果显示,高肺血流性肺动脉高压大鼠血清和肺组织CTRP9水平与其mPAP、mRVP、RVHI、血清NO水平均呈显著正相关(均P<0.05)。结论高肺血流性肺动脉高压大鼠血清和肺组织中存在CTRP9高表达,CTRP9可能参与了高肺血流性肺动脉高压的形成。 Objective To observe the levels of C1q/tumor necrosis factor-related protein 9(CTRP9)in serum and lung tissue of rats with pulmonary hypertension induced by high pulmonary blood flow and its correlation with hemodynamics and right ventricular hypertrophy.Method Fifty adult male SD rats were randomly divided into sham operation group(n=25)and shunt group(n=25).The model of pulmonary hypertension induced by high pulmonary blood flow was made in shunt group.The hemodynamic indexes[mean pulmonary artery pressure(mPAP),mean right ventricular pressure(mRVP)],right ventricular hypertrophy index(RVHI),levels of CTRP9 and nitric oxide(NO)were compared between the two groups.Result The mPAP,mRVP,RVHI,levels of serum NO and CTRP9 in serum and lung tissue of shunt group were significantly higher than those of sham operation group(all P<0.05).Pearson correlation analysis showed that the levels of CTRP9 in serum and lung tissue were positively correlated with mPAP,mRVP,RVHI and serum NO level in rats with pulmonary hypertension induced by high pulmonary blood flow(all P<0.05).Conclusion There is a high expression of CTRP9 in serum and lung tissue of rats with pulmonary hypertension induced by high pulmonary blood flow,CTRP9 may be involved in the formation of pulmonary hypertension induced by high pulmonary blood flow.
