基于在体肠袢模型研究荭草活性成分在正常及心肌缺血模型大鼠的肠吸收特性 被引量:5

Characteristics of intestinal absorption of six active ingredient of Polygonum orientale L.extract in normal and myocardial ischemia model based on intestinal loop model in vivo

摘要 目的基于在体肠袢模型研究荭草提取物(polygonum orientale L.extracts,POE)活性成分在正常及心肌缺血模型大鼠体内的肠吸收特性。方法建立在体肠袢吸收模型,通过肠系膜静脉收集血液,血浆样品经酸化后加甲醇沉淀蛋白,Acquity UPLC BEH C18(2.1×50 mm,1.8μm)柱,流动相:0.1%甲酸乙腈-0.1%甲酸水,流速:0.3 mL·min^-1,采用电喷雾电离源(ESI),扫描方式为多反应离子监测模式(MRM)。结果6个活性成分在正常状态下的吸收随着浓度的升高而增大。心肌缺血模型状态下,原儿茶酸、山柰素-3-O-β-D-葡萄糖苷和槲皮苷的吸收随着浓度的升高而增大,异荭草素、荭草素及牡荆素在中浓度的吸收量小于低浓度及高浓度时的吸收量。6个活性成分正常状态下的主要吸收肠段为十二指肠及回肠,模型状态下的主要吸收肠段为回肠及结肠,且模型组在各个肠段的吸收量均低于正常组。结论荭草提取物中6个活性成分在正常及心肌缺血模型大鼠体内的肠吸收特性存在较大差异。 Aim To study the characteristics of intestinal absorption of six active ingredient of Polygonum orientale L.extract(POE)in normal and myocardial ischemia(MI)model.Methods Intestinal loop model in vivo was established.Blood samples were collected via mesenteric venous.Plasma sample was analyzed by Acquity UPLC C18(2.1×50 mm,1.8μm)column with a mobile phase composed of 0.1%formic acid using step gradient elution after protein deposition and with methanol,and the flow rate was 0.3 mL·min^-1.Quantification was performed on multiple reaction monitoring mode in negative ion modes.Results The absorption of six active ingredients in the normal state increased with the concentration of POE increasing.The absorption of protocatechuic acid,kaempferol-3-O-β-D-glucoside and quercetin increased with the rising of concentration.While the absorption of isoorientin,orientin,vitexin did not display the same changing trend in MI model state.The main absorption segments of the six active ingredients were the duodenum and ileum in the normal state,the ileum and colon in the model state,and the absorption amount in each segment of model group was lower than that of normal group.Conclusion The characteristics of intestinal absorption of six active ingredient of POE in normal and MI model are different.
