摘要
目的探讨基因Nanog和sox2表达在卵巢癌发病、发展与浸润过程中的变化及临床意义。方法选取2013年1月至2017年6月间青岛大学附属医院行手术切除的130例卵巢上皮性肿瘤蜡块标本,术后均经病理确诊,包括卵巢癌80例、卵巢良性肿瘤20例、交界性肿瘤20例和常态卵巢组织10例。采用免疫组织化学方法对卵巢组织中Nanog和Sox2的表达进行检测,分析患者临床病理参数与Sox2和Nanog的相关性。结果Nanog在常态卵巢组织、卵巢上皮性良性、交界性和恶性肿瘤中阳性表达率分别为20.0%(2/10)、20.0%(4/20)、30.0%(6/20)和83.8%(67/80)。Sox2在常态卵巢组织、卵巢上皮性良性、交界性和恶性肿瘤中阳性表达率分别为10.0%(1/10)、25.0%(5/20)、25.0%(5/20)和82.5%(66/80)。Nanog和Sox2在卵巢上皮性恶性肿瘤中呈高表达。在分化程度、临床分期和有无淋巴结转移中,差异均有统计学意义(P<0.05),与病理类型无关(P>0.05)。80例卵巢癌组织标本中,Nanog和Sox2表达均阳性者61例,均阴性者8例。Nanog表达阳性而Sox2表达阴性者6例,Nanog表达阴性而Sox2表达阳性者5例。Spearman级别关联研究(r=0.51,P<0.01),显示Nanog和Sox2在卵巢癌组织中的传达呈现正相关。结论基因Nanog与Sox2在卵巢上皮性癌组织中表达增强,可能预示该肿瘤恶性程度高,侵袭性强、易复发。
Objective To investigate the changes and clinical significance of Nanog and Sox2 in the pathogenesis,development and invasion of ovarian cancer.Methods One hundred and thirty paraffinembedded tissue blocks were collected in patients who underwent surgery at Qingdao University Affiliated Hospital from January 2013 to June 2017 with 80 ovarian cancer tissues,20 benign ovarian tumor tissues,20 borderline ovarian tumor tissues and 10 normal ovarian tissues.The expression of Nanog and Sox2 were detected using immunohistochemistry.The association between patients’clinicopathological parameters and Sox2 and Nanog was analyzed.Results The positive expression rate of Nanog was 20%(2/10),20%(4/20),30%(6/20)and 83.8%(67/80),respectively in normal ovarian tissues,benign ovarian tumor tissues,borderline ovarian tumor tissues and benign ovarian tumor tissues.The positive expression rate of Sox2 was 10%(1/10),25%(5/20),25%(5/20)and 82.5%(66/80),respectively.The expression of Nanog and Sox2 was high in malignant epithelial ovarian tumors.There was significant difference in the expression of Nanog and Sox2 between tissues of different differentiation,clinical stage and with and without lymph node metastasis(all P<0.05).However,it was not associated with pathologic types(P>0.05).Of 80 ovarian cancer tissues,the positive expression of Nanog and Sox2 was observed in 60 and negative expression in 8.Positive expression of Nanog and negative expression of Sox2 was observed in 6 and negative expression of Nanog and positive expression of Sox2 was observed in 5.Spearman’s rank correlation analysis revealed that there was a positive association between clinicopathological parameters and Sox2 and Nanog(P<0.05).Conclusion The enhanced expression of Nanog and Sox2 in epithelial ovarian carcinoma may indicate that the tumor is highly malignant,invasive and prone to recurrence.
